鸡传染性支气管炎病毒基因型与血清型相关性初步研究

摘　　要：为研究鸡传染性支气管炎病毒(IBV)基因型与血清型之间的相关性,本研究以国内不同地区的17株IBV分离株、2株疫苗株(M41、W93)和1株强毒株(X株)为研究对象,经RT-PCR扩增获得20株IBV的S1基因并进行测序。将其分别与GenBank中的20株国内外参考IBV株的S1基因进行序列比较,绘制S1基因系统进化树。结果表明:这20株IBV与20株参考病毒株可分为3个基因型:LX4型、Mass型和X型,其中本研究中绝大多数IBV株属于LX4型。根据这一基因分型结果,从这3个基因型中分别选取差异性较大的6株IBV,即X型的X株,Mass型中的M41、W93、333-3株,LX4型中的DC07-1、333-7株,将其分别免疫SPF鸡制备高免血清,进行血清交叉中和试验,计算IBV各病毒株间抗原性的相关系数(R)。结果表明:6株IBV经血清交叉中和试验可分为3个血清型:333-3、M41、W93株为同一血清型,DC07-1、333-7株为同一血清型,X株为另外一个血清型。以上结果表明:IBV的基因分型与血清分型相一致。这为目前IBV的分型提供一定的参考依据,对今后鸡传染性支气管炎的防控具有一定的意义。To investigate the correlation between genotypes and serotypes of avian infectious bronchitis virus（IBV）,the S1 genes of 17 isolates,two vaccine strains（M41,W93） and one virulent strain of IBV were amplified by RT-PCR and sequenced,which were analysis for genotyping and comparing with those of 20 reference strains in GenBank.The result showed that IBV strains in our study and reference strains were classified into three genotypes of LX4-type,Mass-type and X-type,respectively;most IBV strains belonged to LX4-type.According to the result of genotyping,six highly variable strains from three genotypes,including X strain in X-type,M41,W93,333-3 in Mass-type,DC07-1 and 333-7 in LX4-type,were selected for serotyping by virus neutralization test.Results showed that genotyping of IBV in this study were parallel to the serotypes,which provided references for complicated status of IBV typing currently for the efficient disease control.
