摘  要:目的建立稳定的小鼠微量甲醛炎性痛模型,观察其脊髓背角c-fos的表达规律。方法①16只小鼠随机分成对照组(C组)和炎性痛组(F组),每组8只。F组小鼠左后肢足背注射5%的甲醛溶液3μL,对照组注入等量的甲醛溶液溶酶蒸馏水,分别于注射前,注射后1 h、4 h、12 h、1 d、2 d、3 d、5 d和7 d测定小鼠的热板舔足潜伏期和足趾容积。并记录小鼠在注入甲醛和蒸馏水后0~5 min和20~60min的累积舔足时间,分别代表Ⅰ相和Ⅱ相疼痛程度。②112只小鼠用于c-fos免疫组化测定,112只小鼠随机分成14组,对照组(C1,C2,C3,C4,C5,C6,C7组)和炎性痛组(F1,F2,F3,F4,F5,F6,F7组),每组8只。F组小鼠复制炎性痛模型,对照组不加处理。F组小鼠分别于致炎后1 h、4 h、12 h、1 d、3 d、5d和7 d麻醉处死、灌注后取腰膨大部位的脊髓进行c-fos免疫组化测定。结果①与C组相比,F组小鼠Ⅰ相和Ⅱ相累积舔足时间明显长于C组(P<0.01),F组小鼠与致炎前相比,致炎后各时间点,足明显肿胀(P<0.01),热板舔足潜伏期明显缩短(P<0.01),并出现痛觉过敏。②与C组相比,炎性痛小鼠脊髓中c-fos阳性表达增加(P<0.01)。结论小鼠微量甲醛炎性痛动物模型稳定可行;c-fos可能参与炎性痛的痛觉传导。Objective To establish a stable inflammatory pain model induced by microdosis formaldehyde and to observe the expression of c-fos in neurons of the spinal cord in mice.Methods ① Sixteen mice were randomly divided into 2 groups(n=8),the control group(C group) and the formaldehyde group(F group).The mice in the F group were received a dorsum injection of 3μL 5% formaldehyde solution in the left foot.The mice in the C group were received 3μL distilled water which was the solvent of formaldehyde solution.The paw licking latency and the volume of hind paw were recorded before injection,and at 1h,4h,12h,1d,2d,3d,5d and 7d after injection.And the accumulative licking time in 0~5 min and 20~60 min after the formaldehyde and distilled water injection were recorded,which represent the degree of pain in the phaseⅠ and phaseⅡ.② One hundred and twelve mice were randomly divided into 14 groups(n=8),the control groups including C1~7 group and the formaldehyde group including F1~7 group.The c-fos expression in neurons of the lumbar spinal cord were detected by immunohistochemistry before injection,and at 1h,4h,12h,1d,3d,5d and 7d after injection.Results ①The accumulative licking time of 0~5 min(phaseⅠ) and 20~60 min(phaseⅡ) were longer in F group than those in C group significantly(P0.01).The paw licking latency in F group was shorter than that in C group significantly(P0.01) and the volume of hind paw in F group was larger than that in C group after injection(P0.01),and the hyperalgesia was found in the F group.② Compared with control group,the c-fos expression in neurons of the lumbar spinal cord in the formaldehyde group was upregulated(P0.05).Conclusion The inflammatory pain model in mice induced by microdosis formaldehyde is a feasible and stable model,and the c-fos may play an important role in nociceptive signal pathway related to inflammatory pain.
