摘  要:目的:观察白介素-6(IL-6)在多柔比星肾病大鼠肾组织及尿液中的表达及青藤碱(S IN)对其表达的影响。方法:雄性W istar大鼠120只。随机分为正常组32只,肾病组88只。肾病组大鼠单次尾静脉注射多柔比星5m g/kg,制备微小病变型肾病(M CN S)模型。采用酶联免疫吸附法(EL ISA)检测各组大鼠不同时期肾组织及尿液中IL-6的表达。结果:①肾病组大鼠尿蛋白量逐渐增加,且与正常组比较均有显著性差异(P<0.01);②肾病组大鼠肾组织及尿液中IL-6表达较正常组明显增加,而治疗组较肾病组为低;③肾病组肾组织中及尿液中IL-6的表达与24h尿蛋白量呈正相关(R=0.870,0.794,P<0.01);④肾组织与尿液中IL-6水平也呈正相关(R=0.739,P<0.01)。结论:①肾病组大鼠尿液及肾组织中IL-6含量明显增加,且与24h尿蛋白水平呈正相关,提示IL-6与多柔比星肾病大鼠蛋白尿的发生密切相关;②肾病组大鼠尿液和肾组织中IL-6的水平呈正相关,尿液中IL-6的水平能反映肾组织IL-6的水平;③S IN可能通过下调M CN S中IL-6的表达而减少蛋白尿,从而发挥肾脏保护作用。Objective:To observe the interleukin-6(IL-6) expression in the renal tissue and urine of doxorubicin nephropathy rats and the effect of Sinomenine(SIN) on its expression.Methods: We divided 120 male Wistar rats randomly into normal group(32) and nephropathy group(88).A single intravenous injection of doxorubicin 5mg/kg was given to nephritic rats to prepare with nephropathy(MCNS) model.We used enzyme-linked immunosorbent assay(ELISA) to detect the IL-6 expression in the different periods of the renal tissue and urine.Results:①Nephritic rat urine protein was gradually increased,with a significant difference compared with the normal group(P0.01).②The IL-6 expression in the kidney tissue and urine of the nephropathy group was significantly increased compared with the normal group,while that of the treatment group was lower than that of the nephropathy group.③The IL-6 expression in the kidney tissue and urine of the nephropathy group was positively related to the 24 h urine protein(R=0.870,0.794 P0.01).4.The IL-6 expression level in the kidney tissue was positively correlated with that in the urine(R=0.739,P0.01).Conclusion:①Nephritic rats urine and renal tissue IL-6 levels are significantly increased,positively correlated with the 24h urine protein level,suggesting that IL-6 and doxorubicin nephropathy is closely related to the occurrence of proteinuria.②The IL-6 expression level in the kidney tissue was positively correlated with that in the urine.③SIN could reduce proteinuria by downregulating the IL-6expression in MCNS,which plays a role in kidney protection.
