摘  要:目的:建立测定利肝隆颗粒中黄芪甲苷含量的方法。方法:采用高效液相色谱-蒸发光散射检测器(HPLC-ELSD)法。色谱柱为Agilent C18柱,流动相为乙腈-水(32∶68),流速为1.0mL·min-1,进样量为20μL,柱温为35℃;ELSD检测器漂移管温度为100℃,载气流量为2.7L·min-1。结果:黄芪甲苷进样量在0.3867~3.8670μg范围内与峰面积常用对数的积分值线性关系良好(r=0.9992);加样回收率为99.0%、99.0%、98.7%,RSD=0.6%(n=9)。结论:本方法操作简便、准确、灵敏度高、重复性好,可作为利肝隆颗粒的质量控制方法。OBJECTIVE: To establish the method for the content determination of astragaloside in Liganlong granules. METHODS: HPLC-ELSD method was adopted. The determination was performed on Agilent C18 column with mobile phase consisted of acetonitrile-water (32 ∶ 68) at the flow rate of 1.0 mL·min^-1. The injection volume was 20 μL and column temperature was 35 ℃. The drift tube temperature was 100 ℃ and the flow rate of carrier gas was 2.7 L·min^-1. RESULTS: The linear range of astragaloside was 0.386 7~3.867 0 μg (r=0.999 2). Average recoveries were was 99.0%, 99.0% and 98.7% (RSD=0.6%, n=9). CONCLUSION: The method is simple and accurate, and has good sensitivity and reproducibility. It could be used for quality control of Liganlong granules.
