高效液相色谱法测定三黄散瘀巴布剂中盐酸小檗碱的含量

摘　　要：目的:建立三黄散瘀巴布剂中盐酸小檗碱的HPLC含量测定方法。方法:HPLC法,色谱柱:Agilent Zorbax Eclipse XDB C8(150mm×4.6mm,5μm);流动相:乙腈-0.1%磷酸溶液:50:50(每100mL流动相加入十二烷基硫酸钠0.1g);检测波长:265nm;流速:1mL/min,柱温:25℃;进样量:10μL。结果:盐酸小檗碱在41.2-772.5ng的范围内线性关系良好,回归方程为:Y=2.344X-32.42(r=0.9997);加样回收率在96.91%-102.00%,平均回收率为99.70%,RSD为2.0%。结论:该方法简便快速、结果准确可靠、重复性好,可用于三黄散瘀巴布剂的质量控制。Objective： To establish a content determination method of Berberine hydrochloride in Sanhuang Sanyu Cataplasm by HPLC.Methods： RP-HPLC was performed to determine Berberine hydrochloride on a column of Agilent Zorbax Eclipse XDB-C8（150mm×4.6mm,5μm） at 25℃;Acetonitrile-0.1% H3PO4（50：50）（Adding 0.1g C12H25SO4Na to every 100mL） was used as mobile phase,the flow rate was 1.0mL/min.The detection wavelength was at 265nm.Results： A good linearity can be found in the range of 41.2-772.5ng,and the regression equation was Y= 2.344X-32.42（r=0.9997）.The recovery was between 96.91%-102.0%,and the average recovery rate was 99.70%,RSD was 2.0%.Conclusion： The method was simple,rapid,accurate and available,which can be used to control the quality of Sanhuang Sanyu Cataplasm.
