华蟾素抑制人结肠癌SW-480细胞增殖及诱导凋亡的研究

摘　　要：目的：观察华蟾素（Cinobufacini）抑制人结肠癌（SW-480）细胞系增殖及诱导凋亡的生物学效应。方法：以人结肠癌SW-480细胞为研究对象,采用MTT法观察细胞增殖抑制的影响;采用吖啶橙（AO）/溴乙锭（EB）荧光染色法观察细胞凋亡形态,并计算凋亡率。结果：在20~120μg/mL,华蟾素能明显的抑制SW-480细胞的增殖;AO/EB荧光染色法显示：随着药物浓度的增加细胞凋亡率逐渐增加。结论：华蟾素具有抑制人结肠癌（SW-480）细胞生长增殖,促进凋亡的作用。Objective：To investigate the effect of Cinobufacini on the proliferation inhibition and apoptosis induction in SW-480 Cells.Methods：Human colon cancer cells were used.The growth inhibition of SW-480 Cells was measured by MTT assay.The apoptosis of SW-480 cells was determined by AO/EB fluorescence stain.Results：Cinobufacini exhibited a marked antiproliferative effect on SW-480 cells within the concentration from 20～120 μg/mL by MTT assay.AO/EB fluorescence stain data provided evidence that the apoptosis of SW-480 cells induced by Cinobufacini in a dose-dependent manner.Conclusion：Cinobufacini posseses antitumor effect in vitro.Its mechanism might be associated with inhibition of proliferation and induction of apoptosis.
