摘 要:目的研究糖尿病对骨髓干细胞产生血管生长因子的能力以及向内皮细胞分化能力的影响。方法从糖尿病肥胖大鼠(实验组)和非糖尿病健康大鼠(对照组)各20只分离采集骨髓干细胞进行培养,并检测培养后骨髓干细胞的生存率、培养液中血管内皮细胞生长因子(VEGF)的浓度、以及骨髓干细胞向内皮细胞的分化率。结果实验组经培养3d后骨髓干细胞的生成率为(30.1±2.4)%,与对照组(31.9±3.8)%相比,无显著性差异(P>0.05);实验组经培养7d后骨髓干细胞的生成率为(18.5±3.1)%,与对照组(20.4±1.9)%相比,无显著性差异(P>0.05)。实验组经培养3d后培养基上清液中VEGF的浓度(134.8±27.5)ng.mL-1显著低于对照组(189.1±25.6)ng.mL-1(P<0.01);实验组经培养7d后培养基上清液中VEGF的浓度为(282.7±25.2)ng.mL-1,显著低于对照组(370.2±40.1)ng.mL-1(P<0.01)。实验组经培养7d后骨髓干细胞的内皮细胞分化率为(14.7±4.5)%,显著低于对照组(32.8±5.6)%(P<0.01)。结论糖尿病可以导致骨髓干细胞产生血管生长因子的能力下降以及向内皮细胞分化的能力受损,这将可能导致用自身骨髓干细胞诱导血管新生的能力降低。Objectives To investigate the diabetes-related impair of bone marrow stem cells for inducing angiogenesis.Methods Bone marrow stem cells from 20 diabetic fatty rats(experimental group) and 20 healthy rats(control group) rats were collected and cultured.We measured respectively the cell survival rate of bone marrow stem cells and the concentration of the VEGF in medium after 3 and 7 days of cultivation.Endothelial differentiation rate was also estimated after 7 days of culture.Results The cell survival rate of bone marrow stem cells after 3 days of cultivation from experimental group(30.1±2.4)% and control group(31.9±3.8)% showed no significant difference(P〉0.05).The cell survival rate of bone marrow stem cells after 7 days of cultivation from experimental group(18.5±3.1)% and control group(20.4±1.9)% also showed no significant difference(P〉0.05).The concentration of the VEGF in medium after 3 days of cultivation from experimental group(134.8±27.5)ng·mL^-1 was significantly lower than control group(189.1±25.6)ng·mL^-1(P〈0.01).The concentration of the VEGF in medium after 7 days of cultivation from experimental group(282.7±25.2)ng·mL^-1 was also significantly lower than control group(370.2±40.1)ng·mL^-1(P〈0.01).The endothelial differentiation rate of bone marrow stem cells after 7 days of cultivation from experimental group(14.7±4.5)% was significantly lower than control group(32.8±5.6)%(P〈0.01).Conclusions Bone marrow stem cells form diabetes rats showed significantly decreases of VEGF production and endothelial differentiation,which may indicate diabetes-related impair of angiogenic potency of bone marrow stem cells.
