摘 要:目的探讨miR-138在脊髓全横断大鼠运动功能恢复过程中是否发挥调控作用。方法打开SD大鼠T9-T11椎板,脊髓内分别注射miR-138、DEPC处理水后制备脊髓全横断模型实验组与对照组。术后3d、65d取材,观察脊髓红肿范围、瘢痕长度;期间进行BBB运动功能评分;术后30d应用体感诱发电位(SEP)检测神经功能恢复情况。结果第5~9周miR-138组大鼠BBB评分明显低于对照组(P<0.05)。对照组后肢有运动的大鼠所占比例逐渐增多,而miR-138组该比例未见增加(27.03±16.2,6.5±11.72)(P<0.05)。SEP显示miR-138组大鼠左后肢均未测出潜伏期和波幅,右后肢潜伏期和波幅检出率均为16.7%,明显低于对照组(P<0.05)。术后3dmiR-138组脊髓横断处头端红肿长度较对照组增加(P<0.05)。术后65d脊髓横断处瘢痕明显,与周围软组织及椎骨粘连。miR-138组与对照组比较,瘢痕长度(1.07±0.21,1.03±0.12)cm及周长(1.00±0.17,1.05±0.09)cm均无显著性差异(P>0.05)。结论脊髓全横断大鼠局部应用miR-138后运动功能恢复较对照组缓慢,miR-138在脊髓损伤修复中可能发挥负调控作用。Objective To investigate effects of miRNA-138 on hind limbs in adult rats after spinal cord transsection.Methods Spinal cord injury(SCI) model of SD rats were established by carrying out T9-T11 vertebral plate and transecting spinal cord completely after miR-138 injection into spinal cord near the transsection sites.Gross morphology of spinal cord was observed on 3d and 65d post operation respectively.The motor functional recovery of hind limbs was evaluated once a week for 9 weeks after spinal cord injury by using the open field Basso-Beattie-Bresnahan(BBB) scoring.Somatosensory evoked potential(SEP) of all animals were recorded 30d after injury.Results BBB scores of miR-138 treated rats were significantly lower than those of control group from 5w to 9w(P0.05).The proportion of rats whose BBB scores greater than zero in control group increased gradually,but that of rats in miR-138 group did not increase(P0.05).Latency and amplitude were not detected in the left hind limbs in miR-138 treated rats by SEP inspection.Detectable rate of latency and amplitude in the right hind limb were 16.7%,which were much lower than that of control rats.Range of redness and swelling at the cranial side of spinal cord in miR-138 group were greater than that of control group(P0.05)on 3d post injury.However there was no difference in scar areas between two groups on 65d post operation.Conclusion Motor function of rats receiving miR-138 injection into local spinal cord recovered slowly.miR-138 maybe play a negative regulation role during recovery in spinal cord transected rats.
