通脉大生片对雷公藤致卵巢损伤大鼠卵巢激素与卵泡发育的影响

摘　　要：目的:探究补肾养血法中药复方通脉大生片修复雷公藤多苷致大鼠卵巢功能损害的作用机理。方法:选雌性育龄期SD大鼠行阴道脱落细胞学涂片筛选出动情周期正常者为受试动物,随机选取6只为正常对照组,其余大鼠予雷公藤多苷40 mg/kg.d灌胃造模。取已造模大鼠30只,随机分为5组,每组6只,分别予倍美力等剂量和通脉大生片高、中、低剂量治疗7 d,模型空白组同期予等量生理盐水。各组均于治疗后第8 d采血并处死动物,称重法计算卵巢指数,放射免疫法检测血清雌二醇(E2)、孕酮,酶联免疫吸附法检测抑制素B(INHB);光镜下观察卵巢组织中生长卵泡和黄体数,测量生长卵泡截面积和周长。结果:①雷公藤多苷的生殖毒性使大鼠动情周期紊乱,卵巢指数下降,血中E2、INHB含量明显降低;生长卵泡和黄体数、卵泡截面积和周长均明显减少变小,与正常对照组比较P<0.05。②用通脉大生片治疗后,大鼠动情周期恢复正常,卵巢指数、E2、INHB水平明显升高,卵巢中可见大量初级、次级和接近成熟的卵泡,与模型空白组比较P<0.05,生长卵泡截面积和周长也有升高趋势。结论:①雷公藤多苷可造成育龄期大鼠卵泡发育障碍,补肾养血法复方通脉大生片能增加生长卵泡的数量,提高雌激素和抑制素B水平,从而修复雷公藤所致卵巢损伤,但对孕激素和黄体数量的影响不明显。②补肾养血复方治疗的有效性初步证实该大鼠模型符合中医肾虚证候属性。Objective：To investigate the mechanism involved in the reparative effect of Kidney Qi tonofying and Blood nourishing Chinese medicine compound ＂Tong Mai Da Sheng Pian＂ on the rat model of ovary injury caused by Tripterygium Glycosides.Methods：： Give the vaginal cytology assay to all the SD rats,and filter the laboratory rats which have normal estrous cycle.Take 6 rats randomly to normal control group,and give the others intragastric administration with tripterygium glycosides 40mg/kg/day for 10 weeks.Divide the rats randomly into five groups,6 for each,including the model blank group,the Premarin group,herbal high-dosage group,herbal mid-dosage group,and herbal low-dosage group.Feed them with saline,Premarin and ＂Tong Mai Da Sheng Pian＂ in different dosages separately.At eight days,collect blood and kill the rats.Measure the ovarian index by tissue-weight-assay,check out ＂E2＂,＂P＂ with LEIA,and Inhibin B with ELISA.Observe and count the number of growing follicle and corpus luteum by optical microscope,and measure cross sectional area and circumference of growing follicle.Results：（1） The reproductive toxicity of tripterygium glycosides can cause disorders of estrous cycle,decreasing of ovarian index,E2,INHB,growthing follicles and corpus luteum number,follicle cross sectional area and circumference,P0.05 vs normal control group;（2） ＂Tong Mai Da Sheng Pian＂ has sound effects in curing the ill-conditioning,regulating the estrous cycle,increasing the ovarian index,E2,INHB and growing follicles number,moreover,there＇re large number of primary,secondary follicles and the follicles approaching maturity,And the growing follicle cross sectional area and circumference also has rising tendency,P0.05 vs model blank group.Conclusion：（1） The tripterygium glycosides could cause follicle suppression in rattus norregicus of reproductive age.And the compound ＂Tong Mai Da Sheng Pian＂ pill,which tonifies Kidney Qi and Essence,nourishes Blood,can increase growthing follicles number,INHB
