香连分散片中挥发油的工艺考察

摘　　要：目的:优选香连分散片中木香挥发油水蒸汽蒸馏法的最佳提取工艺,比较N-LOK与β-CD对香连分散片中挥发油的包合能力与工艺过程。方法:以药材饮片的粒径大小、加水量、蒸馏时间、浸泡时间为影响因素,各取3个水平,运用正交实验设计筛选木香挥发油的提取工艺;用研磨法和搅拌法对挥发油进行包合,用DSC,紫外分光光度法,薄层法进行分析。结果:香连分散片中木香挥发油的最佳提取工艺为药材饮片粉碎过3号筛,加8倍量水,浸泡30 min,蒸馏7 h;N-LOK改性淀粉包合效果比β-CD好,最优包合条件为N-LOK改性淀粉与挥发油的比例为4∶1时,采用研磨法制成乳化液再喷雾干燥。结论:筛选的提取工艺操作简单,工艺稳定,挥发油的提取率较高;N-LOK挥发油包合物可直接喷雾干燥,β-CD挥发油包合物需要冷藏及低温干燥。采用N-LOK包合挥发油更接近中成药工业规模生产实际。Objective：To Search for best extracting process in Radix auckladiae in Xiang Lian dispersible tablet;To compare N-LOK with β-CD on inclusion ability for the volatile oi in Radix auckladiae in Xiang Lian dispersible tablet.Methods：The amount of water,distillation time,soak time,particle size of pieces as factors,Select the three levels of each factor to sure the optimal conditions of extracting process in Radix auckladiae by orthogonal experiment design.Grinding and mixing were adapted in the process of including of Radix auckladiae.To analysis the inclusion with DSC,UV spectrophotometry and TLC.Results：The best extracting process was by using 8 times water to immersion the Radix auckladiae which was smashed to through size 3 screen for 30 min,then extracted for 7 h.N-LOK is able to include more oil than the β-CD.We could ge the best effect when the ratio between N-LOK and oil is 4：1.Conclusion： The extracting process can extract essential oil suficiently.And it was stable and easy to operate.N-LOK inclusion complex could spray to dry directly,but β-CD inclusion complex needed refrigeration and low temperaturewere necessary.Themethod of covered with N-LOK is suitable formass production in the traditionalChinese patentmedicine in the size ofmanufacture.
