摘 要:目的 研制四种高危感染菌联合诊断基因芯片并用于临床.方法 通过筛选金黄色葡萄球菌、铜绿假单胞菌、破伤风梭菌及炭疽杆菌的基因序列,获得其有效检测的基因位点;利用已有的基因芯片技术,研制了一张用于四种高危感染菌联合诊断的基因芯片.结果 经临床标本或模拟破伤风梭菌及炭疽杆菌危险品的检测验证,该基因芯片的粗符合率达到93.3%以上,并且检测特异性强.结论 成功制备了四种高危感染菌联合诊断基因芯片,并且具有很好的实用价值.Objective To prepare the combined diagnostic gene chips for four high risk bacteria and used in clinic. Methods The loci of eDNA sequences for valid diagnosis were been identified through the selection of the genome of Staphylococ- cus aureus,Pseudomonas aeruginosa,Clostridium tetani and Anthrax bacillus. The gene chips for diagnosing these bacteria were been developed,based on our own-developed technology for manufacturing and application of gene chips. Results The diagnoses given by such gene chips were consistent well with the reports of clinical laboratories (93.3%) and possess high specificity. Conclusion The corrbined diagnostic gene chips for four high risk bacteria were successfully prepared and well suited for the clinical use in the diagnosis for four high risk bacteria.
