摘  要:对来自福建、浙江和北京地区的中华蜜蜂囊状幼虫病毒(Chinese Sac Brood Virus,CSBV)进行了PCR鉴定,并对来自北京地区的病毒株CSBV-BJ/2010结构基因片段进行了克隆及系统进化分析。结果表明:RT-PCR方法获得了426bp的cDNA片段;所获得序列与GenBank中CSBV-LN/2009病毒株基因序列相似度达90%;序列系统进化分析表明,不同地理来源蜜蜂囊状幼虫病毒(Sac Brood Virus,SBV)可以分为3类,本实验分离得到的CSBV-BJ/2010序列与Isolate A26non structrual gene进化关系较近,而与其他来自欧洲、亚洲的SBV序列同源性差异较大。推测该病毒可能存在着毒力分化。Based on viral RNA structure of Picornaviridae,cDNA structural frament of CSBV genes collected from Zhejiang,Fujian and Beijing were cloned,and phylogeny evolution of SBV genes were analyzed.Results showed that the sequence alignment between a 426 bp-long CSBV-BJ/2010 cloned by RT-PCR and a CSBV-LN/2009 in GenBank is with 90% identities.Furthermore,the evolutionary analysis of SBV genes which had been diverged from 3 groups geographical,revealed that the isolated CSBV fragment has the closest relationship with Sac Brood Virus Isolate A26 Nonstructural protein gene,but has lower sequence homology with SBV genes isolated from other Asian and European strains.The results indicated that a virulent differentiation might be involved in virus Picornaviridae that hosted specially in honey bee.
