摘 要:目的探讨血管性痴呆(VD)小鼠前额叶皮质和海马内磷酸化环磷酸腺苷反应结合蛋白(p-CREB)的变化及其与学习记忆的关系。方法采用双侧颈总动脉线结反复缺血-再灌注法制备多发性脑梗塞性痴呆动物模型;Morris水迷宫测试空间学习记忆改变。免疫组织化学技术以观察小鼠前额叶和海马组织p-CREB的表达变化。结果VD小鼠学习记忆明显受损,海马和前额叶皮质内p-CREB表达明显下降(P〈0.01)。结论VD小鼠学习记忆明显受损,海马和前额皮质中p-CREB可能参与VD小鼠空间学习记忆变化的机制。Objective To detect the expression of phoshorylated cyclic AMP response element binding protein (p-CREB) in the hippocampus and the prefrontal cortex in mice with vascular dementia (VD) and the cognitive ability of VD. Methods The mice were subjected for ischemia and reperfusion repeatedly by the ligation of bilateral common carotid arteries to establish the VD models. The learning and memory of VD mice were investigated by Morris water maze test. Immunohistochemistry was used to observe p-CREB in the hippocampus and the prefrontal cortex. Results The ability of learning and memory in VD mice was worse than that of control group (P〈0.01),and the p-CREB expression in CA3 of hippocampus and prefrontal cortex in VD mice significantly decreased. Conclusion The poor of learning and memory in VD mice were related to the decreased p-CREB expression in the hippocampus and the prefrontal cortex.
