摘 要:目的探讨在4种宫颈癌细胞系中特异表达的水通道蛋白(aquaporins,AQPs)亚型及意义。方法通过水通透性实验比较4种人宫颈癌细胞系SiHa、HeLa、Caski、ME-180的水通透性差异,从中选择水通透性低的细胞系作为实验用细胞系,应用荧光定量PCR、免疫荧光检测细胞系中特异表达的AQPs亚型。结果①SiHa中100 mOsm与300 mOsm、200 mOsm与300 mOsm组间水通透性差异无统计学意义(P>0.05),400mOsm与300 mOsm、500 mOsm与300 mOsm组间水通透性差异有统计学意义(P<0.05);Caski中100 mOsm与300 mOsm、200 mOsm与300 mOsm、500 mOsm与300 mOsm组间水通透性差异有统计学意义(P<0.05),400mOsm与300mOsm组间水通透性差异无统计学意义(P>0.05);HeLa中100 mOsm与300 mOsm、400 mOsm与300 mOsm组间水通透性差异有统计学意义(P<0.05),200 mOsm与300 mOsm、500 mOsm与300 mOsm组间水通透性差异无统计学意义(P>0.05);ME-180中各组间水通透性差异均有统计学意义(P<0.05);②SiHa中表达的特异AQPs亚型有AQP0、AQP1、AQP2、AQP3、AQP4、AQP5和AQP8。结论 SiHa细胞水通透性最低,在SiHa细胞中表达的AQP亚型可能与SiHa细胞的迁移及转移有关。[Objective] To probe and screen isoforms of aquaporins expressing in 4 kinds of cervical eareinoma cell lines. [Methods] Compare osmotic water permeability in SiHa, HeLa, Caski, ME-180 cervical carcinoma cell lines and select carcinoma cell line of low water permeability for the experiment, and examine expression of isoforms of aquaporins by Real-Time PCR and immunofluorescence. [ Results] In SiLa, osmotic water permeability differences between 100 mOsm and 300 mOsm groups, 200 mOsm and 300 mOsm groups are not of statistical significance (P 〉0.05), osmotic water permeability differences between 400 mOsm and 300 mOsm groups, 500 mOsm and 300 mOsm groups are of statistical significance (P 〈0.05); In Caski, osmotic water permeability differences between 100 mOsm and 300 mOsm groups, 200 mOsm and 300 mOsm, 500 mOsm and 300 mOsm groups are of statistical significance (P 〈0.05), osmotic water permeability differences between 400 mOsm and 300 mOsm groups are not of statistical significance (P〉0.05); In HeLa, osmotic waterpermeability differences between 100 mOsm and 300 mOsm groups, 400 mOsm and 300 mOsm groups are of statistical significance (P〈0.05), osmotic water permeability differences between 200 mOsm ancL 300 mOsm, 500 mOsm and 300 mOs'm groups are not of statistical significance (P〉0.05); In ME-180, osmotic water perme- ability differences among groups are of statistical significance (P 〈0.05). AQP0, AQP1, AQP2, AQP3, AQP4, AQP5, AQP8 are isoforms of aquaporins expressing in SiHa cells. [Conclusions] Osmotic water permeability of SiHa cells is the lowest, and isofonns of aquaporins expressing in Siha cells are maybe related with their migration and metastasis.
