摘  要:目的观察丹芪合剂对糖尿病大鼠肾组织Snail1表达的影响,探讨其肾脏保护的可能机制。方法 SD大鼠随机分为正常对照组(A组)、糖尿病组(B组)、丹芪合剂组(C组)和依那普利组(D组)。尾静脉注射链脲佐菌素复制糖尿病模型。生化方法检测尿蛋白、血糖和血肌酐。免疫组织化学法检测肾组织Snail1、E-钙黏素(E-cadherin)、纤维连接蛋白(fibronectin,FN)蛋白水平;反转录-聚合酶链式反应检测Snail1mRNA水平。结果 12周后,药物处理的糖尿病大鼠肾组织Snail1、FN蛋白表达高于正常对照组,但低于未处理的糖尿病组;E-cadherin表达低于正常对照组但高于糖尿病组。Snail1蛋白与E-cadherin蛋白呈显著负相关。Snail1mRNA与其蛋白表达一致。丹芪合剂组与依那普利组相比,Snail1蛋白和mRNA,以及E-cadherin、FN蛋白表达无明显差异。药物组血肌酐和尿蛋白水平低于糖尿病组,但高于正常对照组。结论丹芪合剂可能通过下调Snail1的表达,抑制上皮细胞-间充质转分化(EMT),使FN的生成减少,从而对糖尿病大鼠肾脏起保护作用。[Objective] This study aimed at observing the effects of the Danqi Mixture on the expression of Snaill in the kidney tissue of diabetic rats and exploring the possible mechanism of it's renal protection. [Methods] Sprague-Dawley (SD) rats were divided into normal group(A), diabetic group (B), diabetic group treated with Danqi (C) and Enalapril (D) (n =6). Diabetic rat models were induced using injection of 50 mg/kg streptozotoein (ST'Z). Rats were sacrificed at the week 12. Blood glucose, serum creatinine, and urine protein were examined by biochemistry methods. The protein expression of Snaill, E-Cadherin, FN in renal tissue were measured using immunohistochemistry. The mRNA level of Snaill in renal cortex was examined by RT- PCR. [Results] Comparing to the normal group, the expression of Snaill, FN in renal tissue of diabetic rats treated by Danqi mixture and enalapri increased while the expression of E-cadherin signifieandy decreased. Comparing to DM untreated group, the expression of Snaill and FN in DM treated by Danqi mixture and enalapri decreased and expression of E-Cadherin increased. In group C and D, the levels of Snaill mRNA were down-regulated notably than those in group B. The level of Snaill protein negatively correlated with that of E-eadherin protein in all diabetic rats. The levels of serum creatinine (Set) and urinary albumin excretionin group C and D were alleviated comparing to group B. There were no significant different of Snaill protein or mRNA levels and E-cadherin, FN protein levels between Danqi mixture and enalapri group. [ Conclusion] Danqi mixture could down-regulate Snail1, inhibit the process of EMT and reduce the deposition of FN, there-fore improve injury of diabetic kidney.
