参黄冲剂对衰老患者抗氧化作用及P16基因mRNA表达的影响

摘　　要：目的观察参黄冲剂对衰老患者P16基因mRNA定量表达、血清一氧化氮(NO)和超氧化物歧化酶(SOD)含量的影响,探讨其延缓衰老作用机制。方法将60例衰老患者随机分为参黄冲剂组和对照组,每组30例。在常规疾病治疗基础上,参黄冲剂组加用参黄冲剂,对照组加用甲磺酸双氢麦角毒碱片,均连续服用8周。观察治疗前后患者临床症状及P16基因mRNA定量表达、血清NO和SOD含量变化。结果治疗后参黄冲剂组P16基因mRNA扩增曲线起始循环数明显变大(P<0.05),P16基因mRNA定量表达与对照组比较差异有统计学意义(P<0.05)。参黄冲剂组治疗后血清NO含量和SOD水平明显升高(P<0.05),且明显高于对照组(P<0.05)。参黄冲剂组总有效率为80.0%(24/30),对照组为43.3%(13/30),差异有统计学意义(P<0.01)。结论参黄冲剂可有效改善衰老患者的临床症状,提高日常生活能力,提高血清NO含量和SOD活性,减少P16基因mRNA的表达,其机制可能是通过抑制衰老细胞的增殖而起到延缓衰老的作用。Objective To observe the effect of Shenhuang granule on mRNA expression of P16 gene,level of NO and SOD in serum of senile patients,investigate the its mechanism of anti-aging effect.Methods Sixty cases of senile patients were randomly divided into Shenhuang granule group and control group,30 cases in each group.On the basis of rountine treatment,Shenhuang granule group was treated with Shenhuang granule and the control group with Co-dergocrine Mesylate Tablets additionally for 8 weeks.Clinical symptoms,mRNA expression of P16 gene,serum NO and SOD concentration before and after treatment were observed.Results After treatment,the initial cycle number（ct value） of P16 mRNA gene amplification curve in Shenhuang granule treatment group was significantly largened（P0.05）.P16 mRNA gene quantitative expression had statistical significance between Shenhuang granule group and control group（P0.05）.Serum NO content and SOD level of Shenhuang granule group were significantly increased（P0.05）,and higher than control group（P0.05）.The total effective rate of Shenhuang granule group was 80.0%（24/30）,and control group was 43.3%（13/30）,with significant difference（P0.01）.Conclusion Shenhuang granule can effectively improve clinical symptoms and living ability of senile patients,increase serum NO concentration and SOD activity,decrease mRNA expression of P16 gene.The mechanism is possibly that Shenhuang granule has anti-aging effect by inhibiting senile cell proliferation.
