摘 要:目的以胰岛素抵抗(IR)、雌二醇(E2)、三磷酸甘油醛脱氢酶(GAPDH)为观察指标,探讨菊苣提取物降尿酸的机理。方法鹌鹑按体质量随机分为5组:正常组、模型组、阳性药组及菊苣提取物大、小剂量组,每组15只。正常组给予普通饲料,其余各组均予以造模饲料,同时各给药组予相应药物干预,连续给药28 d。检测血清尿酸(UA)、胰岛素(Ins)、E2水平及全血GAPDH活性,并计算胰岛素抵抗指数(ISI)。结果与正常组比较,用药28 d期间模型组UA水平显著升高(P<0.05)。与模型组比较,菊苣提取物各剂量组UA水平显著降低(P<0.05)。各组比较Ins、ISI均未见显著变化(P>0.05)。与正常组比较,模型组E2水平显著降低(P<0.05)。与模型组比较,菊苣提取物大剂量组E2显著升高(P<0.05)。与正常组比较,模型组GAPDH显著降低(P<0.05),各给药组GAPDH显著升高(P<0.05)。结论菊苣提取物降尿酸的机理可能一方面通过直接升高E2水平促进UA排泄,另一方面还与其升高E2水平,使GAPDH间接升高有关。此外,菊苣提取物还可通过直接升高GAPDH活性降低UA水平。Objective To discuss the mechanism of Cichorii extract in decreasing uric acid regarding insulin resistance(IR) and E2,GAPDH as studying object.Methods Quails were randomly divided into 5 groups by weight:control group,model group,positive contral group(20 mg/kg benzbromarone) and Cichorii extract large and small dosage group(15,10 g/kg respectively),15 quails in each group.The control group was given the common diet,and the other groups were given the yeast powder,continuous administration for 28 days.UA,Ins,E2 level and GAPDH activity were detected,and ISI was calculated.Results Compared with control group,UA level of model group during 28 days was significantly higher.Compared with model group,UA level of Cichorii extract groups were significantly lower.Ins and ISI of each group did not change significantly.Compared with control group,E2 level of model group was significantly lower.Compared with model group,E2 levels of Cichorii extract groups were significantly increased.GAPDH level of model group was significantly lower and Cichorii extract groups were significantly increased.Conclusion The decreasing uric acid mechanism of Cichorii extract is related to increasing E2 level leading to the excretion of uric acid and increasing GAPDH indirectly.Moreover,Cichorii extract can decrease UA level by increasing GAPDH activity.
