银黄方膜分离物对角叉菜胶致大鼠足趾肿胀的影响

摘　　要：目的对采用膜分离工艺得到的各级银黄方膜分离物的抗炎作用进行评价。方法取清洁级雄性Wistar大鼠80只,随机分为10组,每组8只,设正常对照组、模型对照组、阳性对照组、各级银黄方膜分离物组,阳性对照组第1日1 mL/kg、第2日起0.12 mL/kg腹腔注射地塞米松,银黄口服液组按14 mL/kg体质量灌胃银黄口服液,银黄方各组按12 mL/kg体质量灌胃给药,模型组按12 mL/kg灌胃生理盐水,各组均给药6 d,1次/d。第6日各组(正常组除外)均在给药1 h后于右后肢足跖部皮下注射1%的角叉菜胶100μL,造模成功标准以与正常组肿胀度比较有显著差异为准。采用足趾容积测量仪测量造模前及造模后0.5、1、2、3、4、5、6 h右后肢足跖容积,No-Griess法检测6 h时血清及组织液中一氧化氮(NO)水平。结果第三级和第五级膜分离物对角叉菜胶所致大鼠足趾肿胀具有抑制作用,与模型组比较差异有统计学意义(P<0.05)。血清和组织液中NO含量,除第二级膜分离物外,其余膜分离物组均降低,且与模型组差异有统计学意义(P<0.05)。结论银黄方膜分离物对角叉菜胶所致大鼠足趾肿胀急性炎症模型具有抗炎作用。Objective To evaluate the anti-inflammatory effect of membrane separation liquid of Yinhuang decoction.Methods Eighty male Wistar rats（clean level） were divided into 10 groups randomly and each group had 8 rats,including normal group,model group,control group,and various membrane separation liquid of Yinhuang decoction groups.Control（dexamethasone） group was given intraperitoneal injection（first day 1 mL/kg,and 0.12 mL/kg from the next day）,the rest groups（except Yinhuang oral liquid groups according to 14 mL/kg weight） were orally treated according to 12 mL/kg weight for 6 days and once a day.On the sixth day,each group（except the normal group） was injected 1% carrageenan 100 ？L into subcutaneous section of the right hind leg.Successful model criteria was that the swelling degree was significant different from normal group.On the sixth day,the thickness of metatarsus was measured before and 0.5,1,2,3,4,5,6 h after the model was made.Then the level of NO of blood serum and tissue fluid were determined at the sixth hour by No-Griess method.Results All the animals＇ toes swelling caused by carrageenan were reduced in the treating groups that treated with level 3 and level 5 membrane separation liquid of Yinhuang decoction,with significant differences compared with control group（P0.05）.The level of NO of blood serum and tissue fluid in the membrane separation liquid of Yinhuang decoction group,except the level 2,were reduced,with significant differences compared with control group（P0.05）.Conclusion The membrane separation liquid of Yinhuang decoction has anti-inflammatory effect for the toes swelling of rats caused by carrageenan.
