摘 要:目的观察补阳还五汤对脊髓损伤(SCI)大鼠脊髓组织血小板活化因子(PAF)含量的影响并探讨其机制。方法选取SPF级Wistar大鼠,采用Allen’s打击法制作中度SCI模型,参照改良Tarlov评分标准,选择后肢运动功能障碍评分为2~3分的模型大鼠48只,分为模型组、补阳还五汤组、金纳多组。补阳还五汤组给予40 g/kg体质量补阳还五汤水溶液灌胃,金纳多组给予45.5 mg/kg体质量金钠多水溶液灌胃,正常组及模型组给予相同体积的蒸馏水。用药2周后,采用酶联免疫吸附法检测SCI大鼠脊髓组织中PAF含量。结果补阳还五汤组与金纳多组评分随时间变化差异无统计学意义(P>0.05),与模型组及正常组比较,差异有统计学意义(P<0.05)。补阳还五汤及金纳多均可明显降低SCI大鼠脊髓组织PAF含量,与模型组比较差异有统计学意义(P<0.01),且补阳还五汤组明显低于金纳多组,两者比较差异有统计学意义(P<0.01)。结论补阳还五汤改善SCI大鼠后肢运动功能和体质量降低,与其降低脊髓组织PAF含量、减轻PAF对脊髓组织的继发性损伤有关。Objective To explore the influence of Buyang Huanwu Decoction on the content of platelet activating factor(PAF) in spinal cord tissue of the rats with spinal cord injury(SCI).Methods Wistar rats in SPF grade were used for medium SCI models formation.The SCI models were built based on modified Allen′s method.According to modified Tarlov criteria,48 rats with stable condition,whose hind limb motor function evaluation scored 2-3 points,were randomly divided into model blank group,Buyang Huanwu Decoction group and Ginaton group.Rats were treated for 2 weeks with Buyang Huanwu Decoction at a dose of 40 g/(kg·BW) in Buyang Huanwu Decoction group,Ginaton at a dose of 45.5 mg/(kg·BW) in Ginaton group,same volume of distilled water in normal group and model blank group.At the end of the treatment,the content of PAF in the spinal cord tissue of rats was detected by enzyme linked immunosorbent assay(ELISA).Results There was statistical difference between Buyang Huanwu Decoction group and model blank group,normal group(P0.05),but no statistical difference compared with Ginaton group(P0.05).Both Buyang Huanwu Decoction and Ginaton could decrease the content of PAF in the spinal cord tissue of rats with SCI,and the difference between Buyang Huanwu Decoction group and model blank group was statistically significant(P0.01),and Buyang Huanwu Decoction was more efficient than Ginaton with statistical difference(P0.01).Conclusion Buyang Huanwu Decoction can improve the hind limb motor function and inhibit weight loss of rats with SCI,which may be related to the decrease of the PAF content and the relief of the secondary injury of PAF on spinal cord tissue.
