摘 要:采用毛霉菌为菌种,以腺嘌呤为底物转化合成ATP,用Na2HPO4代替反应液中价格较高的K2HPO4,设计单因素实验、正交试验研究其转化条件,初步探索了分批加入Na2HPO4对ATP产率、反应时间的影响.结果表明:转化反应最佳pH为6.5;反应2h后,添加1%的二甲苯,反应2h后,反应液静置2h,可显著提高ATP产率;反应液最佳配方为毛霉菌体500g/L,葡萄糖120g/L,Na2HPO490g/L,MgCl24g/L,MnCl22g/L,腺嘌呤3g/L,二甲苯1%;通过分批加入Na2HPO4使ATP产率由6.218g/L提高至10.046g/L,提高61.56%,反应时间缩短2h.The reactive conditions for ATP production from adenine by Mucor were investigated by single factor experiment. A new reaction liquid using Na2 HPO4 instead of K2 HPO4 (which is more expensive) was proposed and investigated. Based on shaking flask experiments with orthogonal experiment, the optimum reaction liquid was obtained. The effects of stage addition of Na2 HPO4 on ATP yield and reaction time were investigated. As a result, the optimum pH was 6. 5, and the ATP yield was increased remarkably by using 1% xylene at the reaction time of 2 h and placing the reaction liquid staticly for 2 h after the reaction time of 2 h. The optimum reaction liquid was Mucor 500 g/L,glucose 120 g/L,Na2 HPO4 90 g/L,MgCl2 4 g/L,MnCl2 2 g/L,adenine 3 g/L, xylene 1%. The ATP yield was increased from 6. 218 g/L to 10. 046 g/L under the condition of stage addition of Na2HPO4. The yield was increased by 61.56% and the reaction time was decreased by 2 hours.
