摘  要:目的探讨冬虫夏草对四氯化碳致小鼠肝纤维化的作用及肝脏Smad3蛋白表达的影响。方法四氯化碳皮下注射复制BALB/c小鼠肝纤维化模型,虫草组在造模同时予以虫草煎剂,直至造模结束。HE染色观察肝组织炎症,天狼猩红染色观察肝脏胶原沉积,生化法测定肝组织羟脯氨酸(Hyp)含量,免疫组化测定Smad3在肝脏内的表达。结果与正常小鼠比较,肝纤维化小鼠肝脏肝静脉与汇管区周围肝细胞明显脂肪变性,肝脏胶原沉积,可见纤维间隔形成,肝脏Hyp含量、Smad3蛋白表达量均显著增加。与模型组比较,虫草组肝脏炎症与胶原沉积减轻,Smad3蛋白表达量明显降低。结论肝纤维化BALB/c小鼠的肝脏Smad3蛋白表达明显上升,虫草制剂有良好的抗肝纤维化作用,其作用机制与下调Smad3蛋白表达有关。Objective: To explore the anti-fibrosis effect of Cordyceps and its relationship with its regulating the Smad3.Methods:BALB/c mice model of liver fibrosis was replicated by subcutaneous injection of CCl-4.Cordyceps decoction was given to the mice in Cordyceps intervened group when modeling.Hepatic inflammation was observed by HE staining,while hepatic collagen deposition by Sirius red stain.and hepatic hy-droxyproline(Hyp) were determined by biochemical medthod..to observe the histopathologic change in liver and the immunohistochemical staining of Smad3.Results:Compare with the normal mice,obvious fatty dgeneration around hepatic veins and portal area was seen.Hepatic collage deposition and fibrosis septa were seen.Hepatic HYP content and Smad3 protein expression were obviously increased.Compare with the model group,hepatic inflammation and collage deposition were lessened,and Smad3 protein expression obviously decreased in Cordyceps intervened group.Conclusion: Hepatic Smad3 expression obviously increased in,liver fibrosis BALB/c mice.Cordyceps has superior anti-fibrosis.Its mechanism might be through down regulating the Smad3 protein expression.
