摘  要:建立血清中烟酸(NA)、烟酰胺(NM)及其代谢产物N′-甲基尼克酰胺(NMN)、2-吡啶酮(2Py)的测定方法。采用高效液相色谱法(HPLC),以氨基固相柱萃取样品,0.04mol/L KH2PO4提取洗脱,同时测定血清中NMN、NA、2Py、NM的含量。校准曲线线性范围为0.5—30μg/mL,r≥0.9986。检出限为NMN 0.45μg/mL,NA 0.51μg/mL,2Py 0.15μg/mL,NM 0.20μg/mL。回收率为88.6%—105.1%,相对标准偏差(RSD)为1.3%—7.8%。本法简单快速,准确,灵敏,达到了实验目的和要求。The method for the determination of nicotinic acid(NA),nicotinamide(NM),and their metabolites N-methyl-2-pyridone-5-carboxamide(2Py),N′-methylnicotinamide(NMN) in serum was established by high performance liquid chromatography(HPLC).The samples were extracted by amino solid phase column with elution of 0.04mol/L KH2PO4.The contents of NA,NMN,2Py,NM in serum were simultaneously deteceted.The linear range of method was 0.5—30μg/mL with r≥0.9986,and limits of detection were NMN of 0.45μg/mL,NA 0.51μg/mL,2Py 0.15μg/mL,NM 0.20μg/mL,respectively.The recovery was 88.6%—105.1%.The relative standard deviation was 1.3%—7.8%.The method is fast,simple,accurate and sensitive,which meets the experimental purpose and the need.
