血清标本存放时间和存放方式对嗜肺军团菌抗体测定结果的影响

摘　　要：目的探讨血清标本存放时间和存放方式对嗜肺军团菌抗体测定结果的影响,为人群流行病学实际工作中嗜肺军团菌抗体的测定工作提供依据。方法采集120份健康人群血样分为三组,使用酶联免疫吸附试验(ELISA)检测试剂盒,分别在采样当日,以及在室温、冷藏与冷冻条件下保存不同时间后检测,然后进行统计学分析。结果室温组与冷冻组血清抗体LP1-7 IgG在不同存放时间和存放方式条件下定性结果相同,冷藏组除1份阳性标本在第3天测定时落在临界值范围外,其定性结果也无变化。但室温组样品放置24 h、48 h和72 h后定量结果与采样当日测定结果有统计学差异,冷冻组样品放置5周、7周后定量结果与采样当日测定结果有统计学差异。结论血清标本室温保存72 h,在2～8℃冰箱保存5天和-20℃冰箱保存9周对血清中LP1-7 IgG测定定性结果无明显影响,但定量结果不稳定,特别当抗体活性值≥40 U/mL时,波动较大。Objectives To investigate the influence of storage time and storage mode for serum sample on the detection results of Legionella pneumophila antibody, and to provide evidence for the detection of Legionella pneumophila antibody in epidemiological practice. Methods One hundred and twenty blood samples collected from healthy individuals were divided into three groups and detected by ELISA for Legionella pneumophila antibody on the same day of sampling and stored under room temperature, refrigeration and freezing after different days. Results The qualitative results of all stored samples were the same as the results detected on the same day of sampling except one positive sample stored under refrigeration. However, the quantitative results of samples stored for 24, 48 and 72 h under room temperature and stored for 5 weeks and 7 weeks under freezing were significant different from the results detected on the same day of sampling. Conclusions The qualitative detection of Legionella pneumophila antibody for serum samples would not be affected under the storage conditions of 72 h at room temperature, 5 days at 2 -8℃ and 9 weeks at -20℃. However, the quantitative results were not stable under different storage conditions, especially when the activity of antibody was ≥40U/mL.
