摘 要:目的探讨富勒烯(C60)对中国仓鼠肺成纤维(CHL)细胞的氧化损伤作用。方法用不同浓度的C60(0、1.25、2.5、5、10、20和40μg/mL)染毒24 h,四甲基偶氮唑盐实验检测C60对CHL细胞的增殖抑制作用,并检测细胞内LDH释放率及SOD、CAT活性和GSH、MDA、ROS含量。结果 C60染毒组细胞生存率下降,10、20、40μg/mL C60染毒组细胞存活率与对照组相比差异均有统计学意义(P<0.05或P<0.01)。CHL细胞中LDH释放量上升,且各染毒组与对照组相比,差异均有统计学意义(P<0.01)。各染毒组随染毒浓度增加SOD、CAT活性降低,与对照组相比,差异均有统计学意义(P<0.01)。GSH含量降低,但仅C60 40μg/mL染毒组,与对照组比较差异具有统计学意义(P<0.05)。MDA含量增加,10、20、40μg/mL剂量组,与对照组比较差异均具有统计学意义(P<0.01)。ROS含量有不同程度的升高,与对照组相比差异均具有统计学意义(P<0.01)。结论 C60可引起中国仓鼠肺成纤维细胞损伤,可能是通过氧化应激介导。Objectives To study the oxidative damages of fullerenes (C60) on Chinese Hamster Fibroblast cell line ( CHL). Methods CHL cells were treated with C60 of different concentrations for 24 h. Then the effects of C60 on the cells proliferation were measured by monotetrazolium. The activity of LDH, SOD, CAT and the content of GSH, MDA, ROS were determined. Results The results showed that the viability of cells exposed to all groups decreased, and the differences were significant in 10, 20 and 40μg/mL groups (P 〈 0.05 or P 〈 0.01 ). Compared with the control group, the activity of cellular LDH increased in all groups, while SOD and CAT decreased significantly ( P 〈 0.01 ). As to the level of GSH, it was found that only 40μg/mL group had a significant decreased (P 〈 0.05). Meanwhile, the level of MDA increased in 10, 20 and 40 μg/mL group (P 〈0.01) and ROS increased in all groups (P 〈 0.01 ). Conclusions C60 can injure CHL cells. It can be concluded that the oxidative damages could be induced by C60, which leaded to CHL cells injured.
