摘 要:目的探讨围产期低剂量氯化三丁基锡(TBTCl)暴露对子代雄性小鼠发育及血清、睾丸组织中雌激素(E2)和雄激素(T)水平的影响。方法将20只妊娠6 d的SPF级昆明小鼠采用经口灌胃方式进行染毒,剂量为0(对照,玉米油)、1、10、100μg/kg TBTCl溶液,每组5只,染毒至哺乳期结束。仔鼠断乳后每周称量一次体重。出生49天(PND49)时,每组随机抽取10只雄性仔鼠,测定睾丸、附睾、脑、肝、肾、胸腺重量,并计算脏器系数。测定血清和睾丸组织中E2和T水平。结果与对照组相比,除100μg/kg剂量组在PND21、28、35、42、49时体重显著降低(P<0.05,P<0.01)外,其他各组体重均无显著差异;除100μg/kg TBTCl染毒组睾丸系数升高(P<0.01)外,肝脏、脾脏、肾脏、胸腺、附睾的脏器系数均无显著差异;除10及100μg/kg TBTCl染毒组睾丸组织中E2水平降低(P<0.05,P<0.01)外,其他各组血清和睾丸组织中E2和T水平均无显著差异。结论围产期低剂量TBTCl暴露不仅影响雄性小鼠的生长发育,而且降低了睾丸组织中E2水平,并可扰乱小鼠血清、睾丸组织内的雌、雄激素水平的平衡。Objective To explore the effects of perinatal exposure to low-dose tributylin chloride (TBTC1) on development and serum and intratestieular estrogen (E2) and androgen (T) levels in the male offspring mice. Methods Twenty SPF health adult Kunming mice were given TBTC1 at the doses of 0,1,10 and 100μg/(kg-d) respectively by gavage from the 6th gestation day until the end of lactation (corn oil as vehicle, 5 in a group). The pups were weighed once per week after weaning (21 days of age). On postnatal day (PND) 49,10 male offspring mice were randomly selected in each group and the blood was collected. The testis, epididymis, brain,liver, kidneys and the thymus were collected and the viscera coefficients were calculated. The serum and intratestieular E2 and T levels were measured. Results Compared with the control group, the body weight in the 100 μg/kg group significantly dropped on PND21, PND28, PND35, PND42 and PND49 (P〈0.05 ,P〈0.01); the viscera coefficient of the testis (P〈0.01) had a significant increase in 100 txg/kg group. However,there were no significant differences in the gains of body weight,the weight of testis and the viscera coefficient of epididymis,brain,liver,kidneys and the thymus (P〉0.05) between other dose-groups and the control. Intratestieular Ez level in 10 μg/kg and 100 μg/kg groups decreased significantly compared with the control group (P〈0.05,P〈0.01). Conclusion Perinatal exposure to low-dose TBTC1 may not only affect the male offspring mice growth and development, but also reduce the level of E2 in testis, and disturb the homeostasis of E2/T in the male offspring mice serum and testis.
