摘 要:目的探讨维生素E(VE)和维生素C(VC)联合染毒对慢性镉中毒小鼠黑质神经元的保护作用。方法将125只健康3月龄清洁级雄性昆明小鼠按体重随机分为5组,分别为正常对照(生理盐水)组、镉中毒(以2 mg/kg氯化镉溶液经皮下注射染毒,染毒容量为5 ml/kg,每周2次)组、镉中毒+VE(以10 mg/kg VE溶液经口灌胃染毒,染毒容量为10 ml/kg,每天1次)组、镉中毒+VC(以25 mg/kg VC溶液经口灌胃染毒,染毒容量为15 ml/kg,每天1次)组、镉中毒+VE+VC组,每组25只。连续染毒3个月。光镜下观察脑组织黑质区HE染色、尼氏法染色和酪氨酸羟化酶(TH)免疫组化ABC法染色结果,电镜下观察脑组织黑质区域超微结构的改变。采用细胞形态计量方法对黑质区酪氨酸羟化酶(TH)阳性反应神经元进行定量分析。结果 VE和VC单独或联合染毒镉中毒小鼠中脑黑质区结构较镉中毒组有明显改善,且接近于正常对照组。与镉中毒组比较,VE和VC单独或联合染毒镉中毒小鼠脑组织黑质区TH阳性反应神经元细胞核体密度、数密度、平均截面积、平均体积均较高,差异有统计学意义(P<0.05)。各组小鼠脑组织黑质区TH阳性反应神经元细胞核圆球度间比较,差异无统计学意义。对镉中毒的干预效果依次为VE>VE+VC>VC。结论抗氧化剂VE和VC单独或联合染毒对慢性镉中毒组小鼠黑质神经元有明显的保护作用,且以VE的保护效果最佳。Objective To investigate the protective effect of vitamin E (VE) ,vitamin C (VC)on the neurons of suhstantia nigra in mice with chronic cadmium poisoning. Methods A total of 125 male Kunming mice were randomly divided into four groups by weight,25 in each groups. Cadmium poisoning groups and intervention groups were given CdCl2 at 2 mg/kg,through subcutaneous injection,twice a week. VE intervention group was simultaneously administered by VE at 10 mg/kg, through garage once a day, and VC intervention group was simultaneously administered by VC at 25 mg/kg once a day. The combined intervention groups was administered simultaneously by VE at 10 mg/kg plus VC at 25 mg/kg once a day,and the control group was given physiological saline subcutaneously. The histological structure of the substantia was examined,the TH-positive reaction neurons in the substantia nigra were quantitatively analyzed with cell morphometric technique. Results The morphosis of neurons in the substantia nigra of by VE treatment group and VC treatment group and the combined intervention group was similar to the control group,and had significant differences compared with the Cd-exposed group. The protective effect of VE and VC was seen, and the effect ranked as VE 〉VE+VC 〉VC. Conclusion VE and VC may have'protective effect on the neurons of substantia nigra in mice with chronic cadmium poisoning.
