球虫感染对蛋鸡肝脏和肠道mdr_1、mrp_2和bcrp mRNA表达水平的影响

摘　　要：为探索球虫感染对蛋鸡肝脏和肠道组织中多药耐药蛋白基因(mdr1)、多药耐药相关蛋白基因(mrp2)和乳腺癌耐药蛋白基因(bcrp)mRNA表达的影响,采用荧光定量PCR方法,以β-actin为内参,对球虫感染蛋鸡以及健康蛋鸡的肝脏、十二指肠、空肠、回肠中的mdr1和mrp2及bcrp mRNA进行相对定量测定。结果表明:与健康对照鸡比较,球虫感染导致mdr1 mR-NA表达水平在十二指肠中有显著上调(P=0.043),而在其他组织中mRNA表达水平均无显著差异(P>0.05);mrp2 mR-NA表达水平在健康和球虫感染蛋鸡的肝脏及肠道中均无显著差异(P>0.05);bcrp mRNA表达在肝脏(P=0.021)、回肠(P=0.021)、空肠(P=0.021)中均出现显著性下调,在十二指肠中则变化不显著。结论:球虫感染可使蛋鸡组织中的部分ABC(ATP-binding cassette)转运子的mRNA表达水平发生变化,但是不同基因在不同组织的变化趋势不同。提示:疾病期间用药可能会对药物的吸收和排泄有影响。To explore the level changes of ABC（ATP-binding cassette） transporters genes mdr1（multidrug resistance protein 1 gene）,mrp2（multidrug resistance-associated protein 2 gene）and bcrp（breast cancer resistance protein gene）mRNA expression in layers after coccidian infection,real-time PCR method with SYBR Green as dye and β-actin as house-keeping gene was used to quantify the mRNA expression levels of mdr1,mrp2,bcrp in liver,duodenum,ileum and jejunum of coccidian infected chicken′s.The results suggested that with experimentally challenged coccidiosis in layers,mdr1 mRNA expression level in duodenum was up-regulated significantly（P=0.043）,but did not change in other tissues.Changes in the expression level of mrp2 mRNA were not significant.But mRNA expression levels of bcrp in liver（P=0.021）,ileum（P=0.021）and jejunum（P=0.021）were down-regulated significantly,except that in duodenum.The results indicated that mRNA expression levels of some ABC transporters in tissues of layers could be changed due to exprerimentally chanllenged with coccidians but the change level of different genes in each tissue were different.
