摘  要:根据北京鸭线粒体DNA(mtDNA)序列设计引物,对中国9个地方鸭和1个番鸭品种线粒体细胞色素氧化酶Ⅰ(COⅠ)基因序列进行PCR反应和测序,并从起始密码子开始分析了1 520 bp的序列。结果显示:所有家鸭个体在1 517位点出现1个T碱基缺失,番鸭与家鸭的mtDNA COⅠ基因碱基组成相似;9个鸭品种个体共检出11种单倍型,单倍型的多样度为0.856。聚类分析结果显示,所有家鸭品种间的遗传距离为0.001~0.002,种内平均值为0.001;番鸭与家鸭遗传距离为0.003~0.006。基于Kimura-2模型构建的系统发育进化树进行分析,发现所测定的鸭科物种个体大体分为3类:家鸭与栖鸭属的番鸭聚为第1类,应属于鸭亚科;雁属的白额雁、灰雁与天鹅属的黑天鹅聚为第2类,应属于雁亚科;鹊雁与中国骛一起聚为第3类,应属于鹊雁亚科。上述结果表明,基于COⅠ基因条形码,在亚科阶元上,所有鸭科物种分类与以往分类结果是一致的。因此,使用DNA条形码鉴定物种系统发育关系时,需考虑阶元划分标准。In this study,the primers for cytochrome oxidase Ⅰ gene(COⅠ)were designed according to the mitochondrial genome sequence of Peking duck,which were used to amplify a collection of 9 Chinese endemic duck sub-sepecies and Muscovy duck to investigate the phylogenetic relationships among these species.From the beginning code,the CO Ⅰ region of 1 520 bp was used for analysis.The results showed that,there existed only a base T deficiency within 1 517 site.The components of base pair between Chinese endemic duck sub-sepecies and Muscovy duck were similar.There were 11 haplotypes in all Chinese domestic duck,the haplotype diversity(Hd)of which was 0.856.The taxonomy results showed that the genetic distances among Chinese domestic duck breeds were 0.001-0.002,average level being 0.001;the genetic distances between Muscovy duck and Chinese domestic duck breeds were 0.003-0.006.The phylogenetic analysis using the Kimura two-parameter(K2P)mode showed that Anatidae species clustered three groups based on NJ evolution method.The firstly,all Chinese native ducks sequenced in this study and Anas platyrhynchos clustered with Cairina moschata,which belonged to Anatinae;the secondly,Anser albifrons and Anser anser(domestic goose)clustered with Cygnus atratus,which belonged to Anserinae.The last,Anseranas semipalmata(magpie goose)and Egretta eulophotes clustered together,which belonged to Anseranatinae.The results indicated that the composition of all Anatidae species classification were consistent with the previous classified results in the level of order element,which was based on CO Ⅰ DNA barcode approaches.The results suggested that choosing righteous category classification standard is necessary to indentifying phylogenetic relationship via DNA barcode.
