补肾填精中药干预雷公藤多甙致卵巢储备下降大鼠卵巢功能的实验研究

摘　　要：目的:阐释补肾填精中药改善卵巢储备功能的效应机理与靶点。方法:12周龄雌性SD大鼠50只,随机分为5组:空白组、雷公藤模型组、补肾填精中药低、中、高剂量组。观察灌服上述不同药物后大鼠一般情况、卵巢指数、卵巢组织病理形态、血清E2、FSH、P、T水平,卵巢颗粒细胞凋亡因子Bcl-2、Bax、Caspase-3蛋白的表达及血管内皮生长因子VEGF、VEGF受体的表达。结果:补肾填精中药能升高DOR大鼠血清E2水平,降低FSH、P、T水平;上调卵巢颗粒细胞凋亡抑制因子Bcl-2,下调凋亡促进因子Bax、Caspase-3蛋白的表达;对卵巢颗粒细胞VEGF的表达有较弱的促进作用,对VEGF受体的表达无影响。结论:补肾填精中药干预卵巢储备功能低下、保护卵巢的效应机理主要是调节生殖激素、抑制细胞凋亡、促进血管生成,卵巢颗粒细胞是其发挥效应的靶点之一。Objective： To interpret the effect mechanism and targets of tonifying kidney and replenishing essence rescription improve the ovarian reserve.Methods：50 12 week old female SD rats were randomly divided into 5 groups： control group,triptolide group and the group with tonifying kidney and replenishing essence rescription that were divided into high-dosage group,moderate-dose group and low-dose group.After feeding the the different drugs,the rats general situation,ovarian index,pathological morphology of ovarian tissue were observed,serum E2,FSH,P,T levels,expression of ovarian granulosa cell apoptosis factor Bcl-2,Bax,Caspase-3 protein and VEGF,VEGF receptor were detected.Results：The rescription could increase serum E2 levels and lower FSH,P,T levels,It also increased expression of Bcl-2,and reduced expression of Bax,Caspase-3 protein;played a weak role in promoting the expression of VEGF,while it had no effect on VEGF receptor expression.Conclusion：The major effect mechanism of this rescription interfew DOR and protect the ovarian is regulation the hormones,inhibit apoptosis,promote angiogenesis,while the granulosa cells are one of the targets that it works.
