针刺血清配合复方丹参注射液诱导骨髓间充质干细胞向软骨细胞的分化

摘　　要：目的:探讨针刺血清配合丹参注射液对骨髓间充质干细胞(BMSCs)增殖和诱导其向软骨细胞分化的影响。方法:20只SD大鼠电针7d,颈总动脉取血制备血清。30只4周龄SD大鼠骨髓中分离BMSCs,取第3代BMSCs分为空白对照组!针刺血清组、丹参注射液组及针药组,给予相应的诱导条件。结果:MTT显示丹参组及针药组均能促进BMSCs增殖(P<0.05),尤以针药组最为明显,而针刺组与对照组相比差异无显著性(P>0.05)。GAG含量及Ⅱ型胶原免疫组化均显示针药组有显著表达,且较丹参组明显,而针刺组与对照组相比不显著。结论:针刺血清配合复方丹参注射液能有效促进BMSCs增殖和诱导其向软骨细胞分化。Objective：To investigate the effects of acupuncture serum combined with compound Salviae Miltiorrhizae injection on the proliferation of bone marrow mesenchymal stem cells（BMSCs） and inducing to differentiate from BMSCs to chondrocytes.Methods：After 20 SD rats were electroacupunctured for 7 days,blood was sampled from common carotid artery to prepare acupuncture serum.The BMSCs were separated from 30 SD rats＇bone marrow,which were cultivated in vitro.The third passage BMSCs were randomly divided into 4 groups：control group,acupuncture serum group（acupuncture group）,Salviae Miltiorrhizae injection group（drug group） and acupuncture serum combined with Salviae Miltiorrhizae injection group（combined group）.Homologus induction conditions were added to each group.Results：MTT showed that both the combined group and drug group could promote the proliferation of BMSCs（P0.05）.Meanwhile,the effect of the combined group was better than the other groups.Compared to the control group,there was no significant difference between the acupuncture group and control group（P0.05）.The contents of GAG and type Ⅱ collagen of the combined group obviously increased,which were superior to that of the drug group.However,there was no significant difference between the acupuncture group and control group.Conclusion：Acupuncture serum combined with compound Salviae Miltiorrhizae injection can promote BMSCs to proliferate and induce to differentiate from BMSCs to chondrocytes.
