摘 要:目的探讨苦参碱对小鼠成纤维细胞L929的作用及其机制。方法分别应用0.25、0.50、0.75、1.00 g/L苦参碱处理小鼠成纤维细胞L929为24、487、2 h,采用MTT法检测成纤维细胞L929生长抑制率;West-ern blot分析法检测小鼠成纤维细胞L929中磷酸化的eIF4E、eIF4E-BP1和Caspase-3表达。结果在同一作用时间,随着苦参碱浓度的增加,小鼠成纤维细胞L929生长抑制率增加,差异有显著性(F=31.41~41.55,P〈0.01);同一浓度的苦参碱随着作用时间延长,细胞生长抑制率无明显变化。苦参碱作用24 h后,小鼠成纤维细胞L929磷酸化的eIF4E和eIF4E-BP1表达降低,Caspase-3表达增加,差异有显著性(t=9.79~13.32,P〈0.01)。结论苦参碱能诱导小鼠成纤维细胞L929的凋亡,其作用是通过eIF4E、eIF4E-BP1和Caspase-3途径实现的。Objective To study the effect of matrine on fibroblasts L929 in mice and its mechanism. Methods The mouse fibroblast L929 was treated with different concentrations of matrine(0.25,0.50,0.75,and 1.00 g/L) for 24,48 and 72 hours,respectively.The growth suppression rate(GSR) of the L929 cells was measured by MTT assay.Expressions of phospho-eIF4E,phospho-eIF4E-BP1 and Caspase-3 were checked by Western blot analysis. Results At the same action time,the GSR of L929 cells was increased along with the increase of the concentration of matrine(F=31.41-41.55,P0.01).However,the GSR of L929 cells was not changed,at the same concentration of matrine,with the extension of action time.The expressions of phospho-eIF4E and phospho-eIF4E-BP1 were decreased,while the expression of Caspase-3 was increased after 24-hour action of matrine. Conclusion Matrine can induce the apoptosis of L929 cells in mice,which is probably through the ways of eIF4E,eIF4E-BP1 and Caspase-3.
