摘 要:目的采用超高效液相色谱与串联四级杆飞行时间质谱仪联用技术(UPLC/Q-TOF-MS)对厚朴超临界萃取(SFE)提取物进行大鼠体内外化学成分分析。方法用ACQUITY UPLC BEH C18色谱柱,以水(A)/乙腈(B)梯度洗脱,使用ESI离子源,负离子模式下采集数据。初步分析提取物的化学成分、给药后吸收入血成分及其代谢情况。结果厚朴SFE提取物中检测到和厚朴酚、厚朴酚、龙脑基厚朴酚等5个化学成分,其中厚朴酚原型成分吸收入血,并初步鉴定其3个代谢物。结论厚朴酚是厚朴SFE提取物在体内的直接作用物质,结合其代谢产物分析,有助于阐明厚朴SFE提取物的体内药效物质基础。Objective To analyze the consitituents of Cortex Magnoliae Officinalis supercritical fluid extraction(SFE) in vitro and in vivo by using ultra-performance liquid chromatography with quadrupole-time-of-flight mass spectrometry(UPLC/Q-TOF-MS).Methods The chromatographic separation was performed on a ACQUITY UPLC BEH C18 column with a gradient elution of water(A)-acetonitrile(B).The mass spectrometer equipped with electrospray ionization source was used as the detector and was operated under the negative ion modes.The identification of the chemical composition of extracts was performed and metabolism of compound absorbed into blood was also investigated preliminarily.Results Of the 5 original compounds detected,magnolol was absorbed into blood and its 3 metabolites were preliminarily identified.Conclusion Magnolol may be the bioactive component of Cortex Magnoliae Officinalis SFE in vivo.The analysis of its metabolite may lay a foundation for the exploration of the active ingredients of Cortex Magnoliae Officinalis SFE.
