摘　　要：目的进一步研究HCV p7下调基因p7TP2编码蛋白的亚细胞定位。方法构建p7TP2与绿色荧光蛋白(GFP)融合基因的表达载体,转染肝癌细胞系HepG2及绿猴肾细胞系COS-7,通过荧光显微镜观察其亚细胞定位。利用多克隆抗体的免疫组织化学技术,检测p7TP2蛋白在正常肝组织、HBV感染的肝组织、HCV感染的肝组织、肝硬化肝组织、肝癌组织、正常肾脏组织及肾癌组织中的表达。结果蛋白荧光主要集中在细胞浆部分,p7TP2蛋白在阳性组织中同样分布在细胞浆,且在正常组织的表达量显著高于病理组织。结论为进一步p7TP2蛋白的结构和功能的研究奠定了基础。Objective To investigate the subcellular location of p7TP2 protein.Methods A fusion gene of p7TP2 with green fluorescent protein(GFP) was constructed and transfected into HepG2 and COS-7 cell lines,respectively.The subcellular location of GFP-fusion protein was analyzed by fluorescence microscope.p7TP2 expression were detected by immunohistochemistry in normal liver tissue,liver tissue with HBV infection,liver tissue with HCV infection,cirrhotic liver tissue,liver hepatoma tissue,normal kidney tissue and renal carcinoma tissue,respectively.Results The fluorescence of p7TP2-GFP fusion protein was detected mainly in the cytoplasm.p7TP2 protein was also found in cytoplasm of many tissues including normal liver tissue,liver tissue with HBV infection,liver tissue with HCV infection,cirrhotic liver tissue,liver hepatoma tissue,normal kidney tissue and renal carcinoma tissue.It also showed p7TP2 protein expressed higher in normal tissues than that of pathological tissues.Conclusions The study of subcellular location of p7TP2 in cytoplasm established a foundation for its further structural and functional study.
