摘  要:目的观察不同剂量姜黄素对HaCaT细胞中FGF10 mRNA转录及其蛋白翻译的影响。方法HaCaT细胞经不同剂量的姜黄素处理48h后,采用免疫荧光和原位杂交的方法检测FGF10 mRNA转录及其蛋白翻译的改变。结果与未处理组比较,处理48h后,FGF10 mRNA转录及其蛋白翻译水平随姜黄素剂量的增加而减少,两组间比较有显著性差异(P<0.05)。结论姜黄素可抑制HaCaT细胞中FGF10 mRNA转录及其蛋白翻译,并随姜黄素剂量的增加,抑制作用加强。Objective To further illuminate the effect of curcumin on FGF10 mRNA transcription and its protein translation in HaCaT cells.Methods HaCaT cells were treated with different concentration of curcumin for 48h,then the transcription of FGF10 mRNA and the translation of FGF10 protein in these cells were detected by immunofluorescence and in situ hybridization assay.Results Compared with the control groups,with the curcumin concentration increased,FGF10 mRNA transcription and its protein translation levels were decreased.Statistically significant difference was found among different groups(P<0.05).Conclusion Curcumin could have a dose-dependent inhibitory effect on FGF10 mRNA transcription and its protein translation in HaCaT cells.
