摘  要:芥蓝游离小孢子经32℃热激48 h后部分小孢子体积膨大3~4倍,在25℃下继续培养2 d后,可见膨大小孢子进行第1次分裂形成2细胞,4 d后分裂形成4细胞。培养14 d后,只有少部分小孢子可发育成肉眼可见的胚状体,而未膨大或异常膨大的小孢子均不能发育成胚状体。用2个基因型比较了在NLN-13培养基中添加油菜素内脂(BR)对小孢子胚产量的影响,首次发现BR对芥蓝小孢子成胚具有显著的促进作用,其有效浓度范围因基因型有所差异,翠宝的有效浓度为0.005~0.04 mg/L,圆叶则为0.005~0.01 mg/L,两基因型均以浓度0.01 mg/L的效果最佳。In isolated microspore culture of Brassica alboglabra Bailey,some microspores enlarged 3~4 times in size after 48-hour heat shock at 32℃,and their first cell divisions appeared at the second day after transferred to cultivation at 25℃in darkness,and continued to divide to form fourcells at the fourth day.Through two weeks of cultivation,only a few microspores could keep continuous development to form microspore-derived embryos,whereas no embryos developed from non-enlarged microspores or abnormally enlarged ones.Using two genotypes,Cuibao and Yuanye,the embryo yields of isolated microspores were compared in NLN-13 media supplemented with different concentrations of brassionosteride(BR).It was first found that BR had a significant promotion effect on the microspore embryogenesis of Brassica alboglabra Bailey,and the valid concentration differed with genotypes,ranging from 0.05 to 0.04 mg/L for Cuibao and from 0.005 to 0.01 mg/L for Yuanye.The best effect was at 0.01 mg/L for both genotypes
