摘 要:目的采用正交实验设计的方法,提取北豆根中的有效成分———蝙蝠葛碱(M enispermum dauricumDC.)等生物碱,以便优化提取工艺,寻找到一种最优,最高效的提取方法,使之适用于工业生产。方法用正交实验的方法,选定四个因素,每个因素设定三个水平,其中四个因素分别是:硫酸浓度(%);硫酸体积(mL);浸渍时间(h);提取次数,按正交设计表进行提取实验。共进行九次实验,最后称量北豆根总碱的质量,计算收率,并应用药典的方法,即酸碱滴定法,进行含量测定。然后,计算收率和纯度,再折合成生药含量,得出最佳提取工艺。工艺过程:称取北豆根粗粉,用硫酸溶液浸渍,按要求浸渍相应的次数,然后,将所有浸渍液合并,调PH值为碱性,静置6h,吸弃上清液,调底液PH显酸性,静置10m in,进行抽滤,弃去滤液,将滤饼在自然条件下风干即得。结果通过正交数据处理,得出在北豆根总碱提取时,浸渍时间和提取次数对提取的纯度和收率有显著影响,其最佳提取工艺为:0.5%H2SO4,4倍体积,浸渍12h,2次。OBJECTIVE To optimize the extractive process of Menispermum dauricum DC.And fit up to the need of industry preparation.METHODS The orthogonal experiment was used.When these alkaloids were extracted,four factors of this process were considered : The concentration of vitriol(A);the times of extraction(D).There three levels in each factor.According to some experiments,the orthogonal form of four factors and three levels were cited.At last,the content of Menispermum dauricum DC.is measured with the method of pharmacopoeia that is dropping.Dauricine was isolated from the rhizomes of M.dauricum and characterized.The power,rhizome was ext,with 0.5%H2SO4 to give an ext,which was adjusted to PH6.0 and allowed to stand 12 hours.The supernatant then was adjusted to PH9.0,filtrated,and the precipitation was washed with H2O and dried to produce a mix,of crude alkaloids,which were ext.With benzene I was isolated from this mix,by using the standard methods for alkaloid purify. RESULTS The method of the orthogonal experiment showed that.
