摘  要:目的建立反相高效液相色谱法测定人血浆中罗红霉素。方法用乙醚萃取,再用正己烷去除杂质后进HPLC分析,色谱条件为D iamonsil C18柱(5μm,250mm×4.6mm)及D iamonsil C18保护柱;柱温为35℃;流动相为乙腈-甲醇-20mM磷酸二氢钠(0.2%三乙胺,pH 5.5)(37∶18∶45);流速1.0mL.m in-1;紫外检测波长210nm,内标为克拉霉素。结果线性范围0.5~25μg.mL-1,最低检测浓度0.2μg.mL-1,血浆中回收率在99.45~105.0之间,日内、日间RSD在2.33~6.19。结论本法简便、快速,定量准确,适合于人血浆中罗红霉素的测定。OBJECTIVE To develop a method for determination of roxithromycin in human plasma.METHODS Alkalinzed plasma sample was extracted with methylene chloride,and residue was dissolved in methanol-phosphate(pH6.0)(1∶1),and washed with n-hexane.The HPLC system consisted of Diamonsil C18 column(5μm,4.6mm×250mm).The mobile phase was methanol-acetonitrile-20mM(0.2% triethylamine,and adjust pH to 5.5),the flow rate 1.0 mL.min,and the detective wavelength was 210 nm,and clarithromycin was used as internal standard. RESULTS The linear range was 0.5~25μg·mL^(-1),the limit quantification was 0.2μg·mL^(-1),method recovery were within 99.45~105.0,the intra-day RSD and the inter-day RSD were within 2.33~6.19.CONCLUSION This method is simple,rapid and accurate.
