摘 要:目的:提取大鼠腰椎髓核细胞体外培养并观察其细胞生物学特性。方法:显微解剖下分离30日龄SD大鼠的腰椎髓核细胞,体外单层培养,应用MTT检测PDGF-BB和IL-1β处理对培养细胞增殖的影响,同时观察细胞不同时期的超微结构形态学变化。结果:大鼠的腰椎髓核细胞体外单层培养生长扩增良好,没有失去髓核细胞基本生物学特性。结论应用SD大鼠可以建立合格的腰椎髓核细胞体外培养体系。Objective:To cuture the nucleus pulposus cells of the rats in vitro and investiage their biological features Methods:Nucleas pulposus were microsurgically harvested from lumbar Spine of 30 day SD rat. The isolated cells were cultured in monolayer culture system. Cultured cells structure were observated with light and transmission microscopy. The effect of pulposus nucleus cells proliferation were measured by using MTT method. Results were analyzed with standard statisctical techniques. Results:Nucleus pulposus cells proliferated in monolayer culture system Cells maintain their basic biological features, have capacity of responding to biochemical stilmuli such as IL-1β and PDGF-BB. Conclusion:This approach provides a convenient system increase cell number without having to use culture conditions that minic those they may exit in vivo. It is feasible to develop a quantitative nucleus pulposus cells cuture system in vitro without loss of their basic phenotype
