摘  要:在主要固相反应体系中 ,加入少量无毒性的、被德国当局允许用于食品工业的有机溶剂甲乙酮作为反应媒介 ,在釜式膜反应器中经酶催化成功地合成了葡萄糖脂肪酸单酯。为了取得高转化率 ,反应副产品水通过与反应溶剂形成共沸物 ,用共沸蒸馏方式从反应体系中除去。而反应溶剂中的水经过高效分离膜得以除去 ,溶剂得到干燥并循环回流至反应体系中。在实验中 ,运用市售固定化脂肪酶 (ChirazymeL -2 )研究了反应物比、脂肪酸链长对产物得率的影响。在甲乙酮中用超量的硬脂酸反应 ,48h后 ,获得产物最高得率为 90 % ,并发现脂肪酶B (LipaseBCandidaantarctia)对葡萄糖具有高度的选择性 ,在等当量反应物比时 。WT5”BZ]:Production of glucose fatty acid monoesters by enzymatic esterification was carried out in a stirred-tank reactor in a mainly solid phase system in the presence of a very small amount of ethyl methylketone, which is non-toxic and accepted by the German authority for use in the food industry. In order to achieve a high conversion, the water generated during esterification was removed from the reaction medium by azeotropic distillation and was regenerated by membrane vapor permeation. Commercial immobilized lipase (Chirazyme L-2) was used and investigations of the substrate ratio, different fatty acid chain length to influence on the conversion of glucose fatty acid monoesters. Highest yields of fatty acid glucose monoester (90%) was obtained when the reaction was conducted in ethyl methylketone with an excess of stearic acid as acyl donor after 48h. It was found out that lipase B from Candida antarctia has high regioselectivity to glucose and monoacylation was favored at equimolar fatty acid to glucose using long chain fatty acids.
