摘  要:目的用改良的碳酸酐酶(CA)细胞化学方法研究其在肾小管上皮细胞上的分布。方法以铈作捕捉剂将Yokoda的铅-钴置换法改为铈-钴置换法,进行CA的亚细胞定位研究。结果电镜下见铈置换的反应产物颗粒细小,均匀,非特异性沉淀少。观察到CA反应产物主要存在于近曲小管刷状缘质膜,内吞泡膜腔面及线粒体被膜,远曲小管上皮细胞的线粒体被膜上也有分布,但较小。结论铈法及孵育方法的改进获得理想效果,用此细胞化学方法显示的CA,与和表面膜结合的Ⅳ型CA一致,主要功能与近曲小管完成HCO3-重吸收有关。Purpose This work aimed at localization of carbonic anhydrase(CA) on epithelial cells of rat renal tubules using a modified cytochemical method. Methods The cerium cobalt substitution method for CA localization at subcellular level was used to replace Yokoda's lead cobalt method. Results Under the electron microscope,it was observed that the cerium phosphate reaction products(RP) were fine,homogeneous and with less non specific precipitates.The RP deposited on the membrane of microvilli,luminal face of endocytic vesicle membrane and the mitochondrial envelope in proximal tubules.They also deposited on the mitochondrial envelope of distal tubules. Conclusions These findings demonstrate that the cerium based method is simpler and more reliable,and the CA localized by this cytochemical method is concordant with the Ⅳ type CA,which was found at the cell surface membrane and related to the renal reabsorption function of HCO 3 - in proximal tubules.
