满江红茎尖组培苗外观形态及生理生化特性变化

摘  要:采用 0 .1% Hg Cl2 表面消毒 ,剥离 1.0~ 1.5mm大小的茎尖进行组培 ,在 2 5℃ ,50 0 0~70 0 0 lx下培养 30 d便可获得稳定的蕨藻共生的满江红植物体 ,该组培苗植株形态、叠生状态、叶面、叶缘颜色、外观形态和生物量等生物学指标以及固氮酶活性、酯酶同工酶、红萍体内鱼腥藻单克隆抗体等生理生化测定表明 ,它与出发萍体间没有本质差异 ,因此并没有发生变异 ,实验结果表明 ,满江红茎尖组培是一种新的、可靠的品种保存途径。The tissue culture of Azolla tips cut in 1 0-1 5 mm and surface disinfected by 0 1%HgCl 2 was carried out under 5000-7000 lx,25℃ condition The Azolla plants with stable fern algae association were obtained after a 30\|day cultivation The results of observation on appearances including plant morphology,over\|lapped condition,colour of leaf surface and edge,biomass for biological index and N\|fixing activities,esterase isoenzyme,monoclonal antibodies to Anabaena Azolla in Azolla for physiological and biochemical determennation showed that,the plants from Azolla tip culture have no essential variation to compare with the corresponding conventional species The experiments also revealed that the tissue culture for Azolla tips was a new and reliable way to maintain the Azolla germplasm
