用RT-PCR技术诊断猪繁殖与呼吸综合征

摘　　要：利用一对国内自行设计的PCR引物,经反应条件优化,临床样品选择,建立了RT-PCR法,用于猪繁殖与呼吸综合征病原诊断。以模拟临床样品检测,该法敏感度为10TCID50病毒。13个被检猪场中,12个猪场诊断结果与美国IDEXXPRRSV抗体诊断结果相一致。说明该法具有很高的特异性、敏感性和准确性。Through the comparison and selection of the reaction condition and the clinical specimen tested,a reverse transcriptase polymerase chain reaction(RT-PCR)assay was established with a set of primers for the diagnosis of porcine resproductive and respiratory(PRRS).The lowest virus title that could be detected with this method was 10 TCID50.Among 13 clinical ill pig herds tested,twelve had the same results with the RT-PCR as that with ELISA.It showed that this RT PCR had higher specificity and sensitivity for the diagnosis of PRRS.
