摘　　要：以ＨＩＶ－１ ｐ２４ｇａｇ 与ｈＩＬ－６ 基因在重组痘苗病毒中的共表达研究为目的，将痘苗病毒复制非必需区血凝素（ＨＡ） 基因作为侧翼，把编码人白细胞介素６（ｈＩＬ－６）的基因片段克隆到真核表达质粒ｐ１６ＱＦ１ －２４ 的下游，构建成含有ＨＩＶ－ １ｐ２４ｇａｇ 和ｈＩＬ－６ 两种外源基因的重组表达质粒ｐ１６ＱＦ２４ －ＩＬ６ ，经同源重组和血凝素阴性空斑筛选，获得了重组痘苗病毒ｖ１６ＱＦ２４ －ＩＬ６．经间接免疫荧光试验、Ｄｏｔ－ ＥＬＩＳＡ 和Ｗｅｓｔｅｒｎ ｂｏｌｔ 等检测证明，重组病毒能同时表达ｐ２４ｇａｇ 蛋白和ＩＬ－６ 蛋白，表达产物的相对分子质量分别为２－４×１０４ ，２－６×１０４ ．此种方式表达的外源蛋白持续释放，可产生更强的免疫效果，可望用于ＨＩＶ疫苗的研究．The purpose is coexpression of HIV-1 p24gag and IL-6 gene in recombinant vaccinia virus, With flank sequence of HA gene of vaccinia virus ,coexpression plasmid p16QF24-IL6 containing cDNA of human immunodeficiency virus type I envelope gene was constructed by inserting the hIL-2gene under the p16QF1-24 plasmid vector.After homologous recombinantion in Cos-7 cell,recombinant vaccinia virueses were purified in BHK21 cell by using plaque assay. indirect immunofluorescence assay(IFA). Dot ELISA and Western blot showed that the recombinant vaccinia virus had coexpressed the HIV-1 p24gag and IL-6 protein in infected BHK21 and the molecular weight were about 2.4×10 4 and 2.6×10 4.The coexpression of two kinds of proteins can produce strong immune respronse.
