中药复方软坚消肿液抗肿瘤作用及机理研究

摘　　要：探讨软坚消肿液（复方中药）对体外培养的人胃癌细胞凋亡及相关基因表达水平的影响。方法检测细胞凋亡采用DNA缺口末端标记法（TUNEL）、琼脂糖凝胶电泳、透射电镜等技术。原癌基因的表达采用Northern杂交。结果用含软坚消肿液免血清处理的细胞，TUNEL染色、透射电镜均见许多细胞呈现凋亡特征性改变，DNA电泳呈典型梯状图谱；Northern杂交Bc1-2表达较对照组下调，C-myc和P53表达较对照组增强。结论软坚消肿液可诱导MGC-03细胞凋亡，其机理可能是通过调控Bc1-2、C-myc、P53的表达水平实现的，提示软坚消肿液对胃癌防治具有临床应用价值。Objective To explore the effect of the herbal medicinal mixture on apoptosis in human gastric carcinoma cells and level of their correlative genetic expressions. Methods TUNEL, agarose gel electrophoresis and transmission electron microscopy (TEM) were used to detect the apoptosis, and Northern blot to detect the genetic expression. Results Many cells treated with rabbit's serumcontaining the mixture displayed typical changes of apoptosis by TUNEL and TEM. The typical ladderelectrophoresis diagram was displayed in gel electrophoresis. With Northern bl0t, the expression of Bcl-2 was reduced and that of C-myc and P53 was enhanced compared to the controlled group respectively.Concluslon The herbal medicinal mixture is able to induce apoptosis of gastric carcinoma cells, whichmay be by means of controlling the level of Bc1-2, C-myc and Ps,. Our results suggest that the herbalmedicinal mixture has a potential clinical significance.
