人血小板密度亚群异质性的研究

摘　　要：用Percoll间断离心法分离6名健康志愿者血小板密集亚群,研究其与血小板功能关系。结果表明,人血小板存在高、中、低密度(HD、ID、LD)3个亚群。血小板密度、大小、功能及血小板细胞内钙动员量在各亚群明显不同。血小板大小随密度降低而减少(P<0.01)。HD血小板对凝血酶(Thr)、ADP和血清素(5-HT)诱导的聚集反应明显高于LD血小板(P<0.01)。HD对Thr和ADP诱导的ATP释放反应结果也比LD血小板多(P<0.01)。血小板细胞内游离钙对Thr 1500u/L、ADP3μmol/L及5-HT3μmol/L诱导的钙动员量也随血小板密度增高而增加。证明人血小板存在异质性,高密度血小板功能及活性均高于LD血小板。Using percoll discontinuous gradient prepared platlets density subpopulations from six healthy volunteers, to study the relationship between platelet density and the functions. The data indicated that human platelets were separated into high density(HD), intermediate density(ID) and low density(LD) subpopulations. The platlet density ,size,functious and the cytosolic- free calcium mobilization were different in platelet density sub-populations, and their size were diminished with decreasing density(P<0. 01 ). The magnitude of aggregation by thrombin(Thr) ,ADP and 5-HT was more signifcant in HD Platlets than that in LD(P<0.01). The amount of secretion of ATP induced by Thr and ADP in LD(P<0.01). Thr(1500u/L) ,ADP 3μmol/L and 5 - HT(3μmol/ L) induced cyto - solic - free calcium mobilization was heightened with increasing density. The present work suggested that the existence of a heterogeneity in human platelet and the function of HD platelets was much stronger and move active than that of LD.
