摘  要:目的:探索并建立低温保存血小板实验方法和技术,以解决临床用血小板难的问题。方法:以终浓度为4%~5%DMSO,于—80℃条件下保存从新鲜血液中分离提取的、仍保持较高活性的血小板为对象,进行低温生物学研究,并做了853例患者输注临床观察。结果:血小板保存6~16个月,采用联合诱导剂检测血小板功能,其血小板聚集功能和粘附率保持在新鲜血小板水平的90%以上,低渗休克反应恢复率为52%±10.1%,KPTT、Ⅶ因子R:Ag及正因子有效性亦保持正常,临床输注后血小板数明显升高,获得满意的止血效果。结论:DMSO系细胞内超低温保护剂,于—80℃条件下仍能有效地保存血小板活性;输注含有DMSO的血小板,对人体是安全的,并具有提高血小板的作用和良好的止血功能。Purpose:experiments were performed in order to solve the problems in the long term cyopreserved platelets can be available to the patients. Methods: The platelets separated from fresh blood of 853 cases were frozen at -80℃ in freezer immediately at the final concentration of 4 %~5 % DMSO. Result: 6~16 months after cryopreservation, the aggregation function of platelets were tested with combined reagents(inducer). Thier level of aggregation and adhension rate were 90% higher than those of fresh platelets. The results of KPTT,the activity of faster Ⅷ and factor Ⅲ were all at the normal range. After infusion of these cryopreserved platelets, the platelet count of patients increased markedly and hemorrhage events were all controlled effectively. Conclusion: DMSO is a kind of intracellular cryoprotectent, which can maintain to activity of cryopreseve platelet at -80℃. Infusion of cryopreserved platelet containing only a litte amout of DMSO will not do obviously harm to human body,the function of hemostasis was observed.
