摘 要:目的:探讨内皮素1(ET-1)对烧伤后血管通透性的影响。方法:大鼠30%体表面积Ⅲ度烫伤后,用放射免疫分析法测定血浆ET-1的浓度变化;烫伤后即刻及1,2,3h分别静注ET-1特异性抗血清(滴度1:5000)1ml/kg,观察烫伤大鼠平均动脉压和[125I]-白蛋白分布的变化。结果:大鼠烫伤后,血浆ET-1浓度很快升高,伤后4h达高峰,24h仍明显高于正常对照,48h降至正常水平。延迟复苏大鼠的ET-1浓度,虽然与立即复苏组没有显著差别,但各时相点的ET-1浓度均高于立即复苏组。静注ET-1抗血清组的大鼠平均动脉压与对照组无显著差别,但[125I]-白蛋白在心、肺、空肠和骨胳肌组织的分布明显少于对照组。结论:烧伤后血浆ET-1参与烧伤后血管通透性增加的发生发展。Objective:To investigate the pathogenetic role of endothelin-1 (ET-1) in burn shock. Methods:The plasma ET-1 concentration in rats with 30% third degree burn was investigated by radioim munoassay. and following injection of ET-1 antiserum immediately and at 1 , 2, 3 h postburn,the mean arterial pressure(MAP) and distribution of [125I]-albumin in burned rats were measured. Results: The plasma ET-1 concentration in burned rats increased further,with its peak at 4 h postburn. Thereafter ETl concentration decreased but still increased at 24 h , and returned to baseline level at 48 h postburn. Every value was higher in the delayed resuscitation group than that in the immediate resuscitation group. although there was no significant difference within the two groups. Injection of ET-1 antiserum had no marked effect on MAP. but significantly decreased the distribution of [125I]-albumin in the heart . lung,jejunum and muscle in burned rats. Conclusion: Plasma ET-1 level is one of the important pathogenetic factors of vascular permeability enhancement postburn.
