葆春丸和丹参对老龄大鼠血小板与动脉花生四烯酸代谢的影响

摘  要:测定不同月龄大鼠血小板最大聚集率(MAR)、花生四烯酸(AA)诱导血小板的血栓素 B_2(TXB_2)和动脉组织6-酮-前列腺素 F_(1α)(6-keto-PGF_(1α))、TXB_2含量及动脉前列环素(PGI_2)样活性,并观察葆春丸和丹参片对老龄大鼠上述指标的作用。结果表明:老龄对照组MAR 和 AA 诱导血小板 TXB_2量明显高于青年组,葆春丸组和丹参片组这两项指标显著低于对照组,并接近于青年组水平。老龄对照组动脉 PGI_2样活性和 AA 诱导动脉壁的6-keto-PGF_(1α)与6-keto-PGF_(1α)/TXB_2比值较青年组显著低下,两用药组 PGI_2样活性和葆春丸组6-keto-PGF_(1α)/TXB_2比值均明显高于对照组,并与青年组水平接近。提示:老龄大鼠血小板 AA代谢增强和动脉组织 AA 代谢降低及动脉抑制血小板活化的作用低下,葆春丸和丹参片对上述变化有不同程度的改善作用,葆春丸的作用较为广泛,这可能是葆春丸抑制血小板功能随增龄而增强的主要机制之一。It was studied that effect of Baochun Pill(BCP)and Danshen tablet(DST)on maximum platelet aggregation ratio (MAR),TX_2 in platelets and 6-keto-PGF_(1α),TXB2 in aortic tissue induced by araehidonic acid(AA)and PGI_2-like ac- tivity in the arotic tissue of aged rats.The results showed that the MAR and TXB_2 in the platelet were higher,6-keto- PGF_(1α)and 6-keto-PGF_(1α)/TXP_2 as well as PGI_2-1ike activity in aortic tissue were significantly decreased in the control group(21 months old)and than those in the younger(5 months old).The reduction of MAR and TXB_2 and the elevation of PGI_2-like activity in BCP's(21 months old)and DST's(21 months old)were significantly obvious than those in control group.The 6-keto-PGF_(1α)/TXB_2 in aortic tissue was enhanced in BCP's compared to control group.It was suggested that the AA metabolism increased in platelet and decreased in aortic tissue in aged rat,that BCP's effects on inhibiting platelet TXA_2 and enhancing aortic tissue 6-keto-PGF_(1α)/TXB_2 induced by AA as well as increasing PGI_2-like activity were signifi- cant,it might be one of the mechanisms of BCP against thrombopoiesis in the aged.
