承气合剂等对内毒素体外诱生肿瘤坏死因子的影响

摘　　要：为明确承气合剂、庆大霉素、氢化考的松及大肠杆菌内毒素体外对小鼠腹腔巨噬细胞分泌肿瘤坏死因子(TNF)的影响。本实验采用细胞培养及细胞毒生物测定法,分别观察了各因素(单独或组合后)对细胞培养上清中 TNF 细胞毒性的诱生作用。结果表明,经内毒素(LPS)刺激后,细胞培养上清对靶细胞具有强烈的细胞毒性。庆大霉素也表现出一定的诱生 TNF 作用。当各因素分别与 LPS 配伍后发现,低浓度的氢化考的松便可对 LPS刺激巨噬细胞分泌 TNF 产生抑制效应。浓度增高后承气合剂也表现出一定的抑制效应。承气合剂与氢化考的松合用可能具有协同抑制作用。In order to study the effect of Chengqi mixture(CQM),Gentamicin and Hydrocortisone on experimentally lipopolysaccharide(LPS)-induced tumor necrosis factor(TNF)secretion,the murine L929 fibroblast bioassay was employed to observe the changs of TNF cytotoxicity in culture supernatant of mice's peritoneal macrophages stimulated by LPS, which was co-cultured with one drug or the comb ination of drugs.The supernatant of macrophages stimulated bv LPS of different concent rations demonstrated strong cytotoxicity to L929 cell line(P<0.01).Gentamicin of different concentra- tions also played a role in inducing TNF cytotoxicity to varying degrees(P<0.05).After combining several drugs with LPS separately,it is found thai hydrocortisone of low concentration showed an inhibitive effect on LPS-induced TNF se- cretion and the effect gradually became obvious by raising its concentration(P<0.01).Although CQM of low concentra- tion did not have inhibitive effect,as the concentration was raised it also showed an inhibitive effect in some extent(P< 0.05).it was also found that the combination of CQM,hydrocortisone and LPS in low concentration could weaken the l.PS-induced TNF cytotoxicity obviously(P<0.01).But the exact mechanisms of the above effects still need to be ex- plored further.
