蛋白负荷对实验性慢性肾功能衰竭大鼠肾损伤的作用及机理研究

摘  要:本研究观察了不同蛋白负荷对腺嘌吟所致慢性肾功能衰竭(CRF)大鼠产生慢性刺激后,肾比重、肾脏超微结构、血尿素氮(BUN)、血肌酐(SCr)、24h 尿蛋白量(UP)、血胆固醇(TC)、血甘油三酯(TG)、过氧化脂质(LPO)、超氧化物歧化酶(SOD)、维生素 E(Vit E)、谷胱甘肽过氧化物酶(GSH-Px)等的变化。结果表明,高蛋白饮食能明显加重肾小管和间质的损伤,肾比重增加,LPO、BUN、SCr、UP、TC、TG 明显增加(P 均<0.01),而SOD、Vit E、GSH-Px 显著降低(P<0.01,0.05,0.01)。提示,高蛋白饮食能加重肾损伤,加速 CRF 病程进展,是各种因素的变异综合作用所致。The present study investigated the effects of various protein dietary load on antioxidation and renal function as well as uhrastructure in animal model of adenine-induced chronic renal failure in rats.The results showed that high protein diet enhanced significantly the pathological changes of renal tubular and interstitium which has been injured by adenine,in- creased the activity of superoxide dismutase in erythrocytes(P<0.01),the activity of glutathion peroxidase in blood(P <0.01)and the content of serum vitamine E(P<0.05).It also decreased remarkably the content of lipid peroxide in serum(P<0.01).Blood urea nitrogen,serum creatinine and proteinuria were reduced significantly(P<0.01,P<0. 01,P<0.01).These results suggested that high protein diet play an important role in accelerating the progression of chronic renal failure.
